Mycelial Growth of Pleurotus Citrinopileatus on Different Environmental Condition

Abstract/ Executive Summary:

The effects of culture media, pH, temperature, carbon and nitrogen source, C/N ratio and light intensity on growth of Pleurotus citrinopileatus were investigated. This mushroom produced best mycelial colony (0.88cm/day) on corn extract agar medium followed by malt extract agar medium (0.57 cm/day) after incubation. In case of corn extract agar, mycelial colony was found highest but mycelial density was very thin compared to malt extract agar and best mycelial growth was observed at 25-27 ^oC while no mycelial growth was produced at >35 ^oC or <10 ^oC. Likewise, very good growth was produced at pH 8.0 after 7 days of incubation. Among the carbohydrate sources tested, sucrose and maltose stimulated highest mycelial yield of 0.29 cm/day. All the nitrogen compounds investigated enhanced significant mycelial production. However, yeast stimulated the best mycelial yield (0.34 cm/day) followed by peptone (0.33cm/day). Carbon to nitrogen ratio of 12: 1 stim ulated good mycelial growth and highest mycelial growth (0.54 cm/day) was also found in absence of light (0 lux) condition.

Objectives:

To investigate the effects of different media, pH level, light intensity, temperature, carbon source, nitrogen source and C:N ratio on vegetative growth of this mushroom.

Methodology:

The experiment was conducted in the tissue culture laboratory of National Mushroom Development and Extension Centre (NAMDEC) during July to October 2008. Different synthetic media such as potato dextrose agar (PDA), wheat extract agar (WEA), malt extract agar (MEA), malt yeast extract agar (MYA), corn extract agar (CEA), cane juice agar (CJA) and yeast extract agar (YEA), were used in this study. The basal components of each media were mixed with 20g/l of agar separately. Then the mixtures were boiled on gas burner until the agar dissolved. The media were adjusted pH value at 6.5 before poured into petri dishes (90 mm diameter) at 20 ml/plates and sterilized in an autoclave for 20 minutes at 121 ^oC and 1 kg/cm 2 pressure. After sterilization and solidification, the plates were inoculated with the inocula of Pleurotus citrinopileatus mushroom. Then plates were transferred into incubation room for mycelium running at 25 ±2 ^oC temperature. Average mycelial growth and duration of complete mycelium running of each replication was calculated. Plates of malt extract agar medium (MEA), adjusted to pH 4.0, 4.5, 5.0, 5.5, 6.0, 6.5, 7.0, 7.5, 8.0 and 9.0 were inoculated as above and incubated at 25 ± 2 ^oC . The selected medium was employed to evaluate the suitable tempareture and light for mycelial growth of Pleurotus citrinopileatus. The inoculated plates were incubated at five different ranges of temperature values viz: 8-10 ^oC, 12-15 ^oC, 20 ^oC, 25-27 ^oC, 28-30 ^oC, 35 ^oC and 40 ^oC respectively. In case of light, the inoculated plates were placed at five different light conditions viz: continuous dark (0 lux), 40- 60 lux, 100-130 lux, 250-300 lux and alternation of light and darkness (12 hrs light and 12 hrs dark) separately for incubated at 25 ± 2 ^oC. Average mycelial growth of each replication was calculated. Carbon sources selected were dextrose, sucrose, maltose, fructose and lactose. 10 gram each of the carbon sources wase incorporated into the basal medium and distilled water added to make 1 litre. The following nitrogen compound was selected- sodium nitrate (NaNO₃), ammonium nitrate (NH₄NO₃), peptone, yeast extract and urea. Each were added 20 g to the basal medium and made up to 1 litre with distilled water. Sterilization, inoculation and incubation were carried out as described above. Based on the chemical composition of base medium (30 g glucose, 4 g yeast extract per litre of distilled water) seven media with different ratios (6:1, 12:1, 18:1, 24:1, 36:1, 48:1 and 96:1) were prepared. Sterilization, inoculation and incubation were carried out as described above.

Achievement/Findings:

The present study states that conditions required for best mycelial growth of the Pleurotus citrinopileatus include corn extract agar (CEA) medium, temperature range 25-27 ^oC, pH-8.0, sucrose and maltose as carbon source, yeast extract as nitrogen source, 12: 1 as the carbon to nitrogen ratio and in absence of light (0 lux) condition.
