In vitro culture conditions such as media, pH, temperature, light, media concentrations, carbon and nitrogen sources were investigated for mycelial growth of Coriolus versicolor. Among the ten different culture media, the highest mycelial growth was obtained in malt extract agar medium followed by potato dextrose agar and wheat extract agar media while the lowest mycelial growth was observed in yeast extract agar medium. The optimum mycelial growth was found at the pH 5.5 followed by 6.0, 6.5 and 7.0, respectively. The optimal temperature for mycelial growth was recorded at 29-31 oC whereas poor mycelial growth was recorded at 35 oC. In respect of availability of light, 12 hours alternation of 250 and 0 lux was found better than continuous 250-300 lux and continuous dark. The different concentrations of malt extract influenced the growth of mycelium. The highest mycelial growth was found in 40 g/l to 50 g/l of malt. The most suitable carbon source for the mycelial growth was sucrose followed by D-glucose while the least was found with maltose. As source of nitrogen, peptone was found superior to yeast and sodium nitrate whereas urea hampered the mycelial growth of Coriolus versicolor.

To determine the optimal in vitro cultural conditions of the mycelial growth of Coriolus versicolor.

The experiment was conducted in the tissue culture laboratory of National Mushroom Development and Extension Centre (NAMDEC) Savar, Dhaka during August to November, 2008 to examine the cultural conditions for mycelial growth of Coriolus versicolor. The following procedures were maintained in every step. Ten different culture media such as potato dextrose agar medium, potato dextrose yeast agar medium, malt extract agar medium, malt extract yeast agar medium, yeast extract agar medium, wheat extract agar medium, wheat extract yeast agar medium, corn extract agar medium, corn extract yeast agar medium and sawdust extract agar medium were used to determine the best medium for mycelial growth of C. versicolor. From the previous experiment the best medium sorted out was employed to screen the suitable pH value for mycelial growth of Coriolus versicolor. Same composition was used to prepare the selected medium. The medium was adjusted to different pH values at 5.0, 5.5, 6.0, 6.5, 7.0, 7.5, 8.0, 8.5 and 9.0 using 1N HCl or 1N NaOH before those were autoclaved. After cooling the Petri plates were inoculated with the inocula of Coriolus versicolor. Then the plates were transferred into incubation room for mycelium running at 25±2 oC temperature. Average mycelial growth of each replication was calculated as mentioned above. The medium was adjusted at pH value 6.5 before autoclaved. After cooling the Petri plates were inoculated with the inocula of Coriolus versicolor . Then the inoculated plates were incubated at five different temperature ranges viz. 13-15 oC, 20-22 oC, 25-27 oC, 29-31oC and 35oC. Different concentrations of malt extract on mycelial growth were investigated in malt extract agar medium. Five different concentrations such as 10 g/l, 20 g/l, 30 g/l, 40 g/l and 50 g/l were used in this study. The medium was adjusted at pH value 6.5 before autoclaved. After inoculation the plates were transferred into incubation room for mycelium growth. Average mycelial growth of each replication was calculated in the same process as described earlier. Five different carbon sources: sucrose, fructose, lactose, maltose and D-glucose and nitrogen sources: peptone, yeast, sodium nitrate, ammonium nitrate and urea were used to examine the response of mycelial growth of Coriolus versicolor. The liquid medium was prepared separately at the rate of 20g per litre of each source of carbon and nitrogen compound. The experiment was laid out following completely randomized design (CRD) with four replications. Data on mycelium growth rate (cm/day) and duration to complete mycelium running (days) were recorded. Data were analyzed following MSTST-c computer program. Means were compared following Duncan’s Multiple Range Test (DMRT) using the same computer program.

Thus it may be concluded that for optimal mycelial growth of Coriolus versicolor the best medium was malt extract agar with 40g/l malt at pH 5.5. The optimal temperature and light requirements were 29-31 oC and 250 and 0 lux (at 12 hours alternate cycle of light and darkness). The best source of carbon and nitrogen was found sucrose and peptone.
