Screening of Wheat Germplasms Against Some Isolates of Heliminthosphrium Sativum

Abstract/ Executive Summary:

Helminthosporium sativum is one of the most virulent pathogens in wheat, Wide variation in respect of virulence among the isolates of H. sativum was found. Wheat germplasms/lines/cultivars varied from one another in response to their disease reaction against the different isolates of pathogen. Moreover, wheat materials showed different response to pathogen at two different plant growth stages. Plants were found to be more susceptible at seedling stage than that of at flag leaf stage. Few materials were graded to be resistant, but none was found to be highly resistant.

Objectives:

The present research programme was undertaken to evaluate the reaction of 60 wheat materials to some isolates of H. setivum by artificial inoculation method at seedling stage and flag leaf stage.

Methodology:

Inoculum collection and meneqement :The isolates of H. sativum were collected from wheat leaves from different locations of Bangladesh in 1989-90 and were maintained on sterilized wheat grains and were stored in refrigerator at 4°C (Gilchrist 1984). Inoculum increase and its preparation: Desired amount of inoculum was prepared by transferring infected grains to petridishes with PDA and was incubated at 22-4°C for a week. Later the spore suspension was prepared to 60000 spores/mI following the CIMMYT method for screening wheat for H. sativum resistance (Gilchrist 1984). Preparation of test material and inoculation: The experiment was conducted in two growth staqe of plants. Firstly at seedling stage and secondly at flag leaf stage. In case of seedling stage inoculation test 50 seeds of each material for testing each Isolate of the pathogen were sown in plastic pots (20 cm dia.) containing soil and compost (4:1). The plants at 3-4 leaf stage were sprayed with the spore suspension. The seedlings were placed into a room at 22- 24°C in darkness for 24 hrs. where 95% relative humidity was maintained. The inoculated plants then transferred into a room under normal light (12/12) at 22/24^oC having 80% relative humidity. After six days the plants were evaluated following the rating scale of Alam and Gustafson (1983). In case of flag leaf stage inoculation, 15 seeds of each material for each isolate of the pathogen were sown in plastic pots (20 cm dia.) containing soiI and compost (4: 1) in triplicates. The plants were inoculated and incubate d as don e at seedling stage and were evaluated following the modified rating scale of Gilchnst (1984) and Alam and Gustafson (1988) as used for scoring the plants under field condition by Hossain (1991).

Achievement/Findings:

The present study showed that about six materials have shown resistant reaction against all isolates used at flag leaf stage. These wheat materials need to be tested further for their response to other isolates before being used as a resistant source. Moreover, these materials required to be tested in the field condition under sufficient inoculum potential.
