Further Studies on the Improvement of Propagation Techniques of Mango (m Angifera Indica L.).

Abstract/ Executive Summary:

Experiments were carried out for modification in the procedure of veneer grafting and the development of micropropagation technique for the improvement of mango propagation systems. Veneer grafting on medium-soft, medium-hard and hard-woody stem showed that grafting between younger partners is more affective (having 98% success) than that of hard-woody stem of normal practice. Based on this result, veneer grafting on younger internodes was practiced for the production of different varieties of mango on the same plant. This was done on short-sized plants for the production of good quality fruits and on tall plants for both good quality timbers and fruits. An encouraging result with 88% success was obtained in this study. For micropropagation technique, 0.1% HgCl₂ was found to be effective in initial experiments on decontamination of shoot tips of mango among the three sterilants, NaOCI₂, HgCl₂ and H₂0₂, available locally.

Objectives:

A view to (i) find out the youngest stage of mango stem suitable for the formation of effective graft union, (ii) modify and improve the procedure of veneer grafting of mango for its wide application and (iii) develop rnicropropagation technique for the production of both quality timbers and fruits as well as for varietal improvement.

Methodology:

The experiment was carried out in 1991 on approximately one and two years old plants grown from stones of a single tree of local type in June of 1989 and 1990 at the Agroforestry Germplasm Centre, Department of Crop Botany. BAU. Mymensingh. Seedlings were grown in rows of 0.5 m apart in blocks of 7 m x 3 m each. Grafting operations were done in the months of May, June, July and August of 1991 on three stages of stem. These are medium-soft internodes of tip regions (1st or 2nd inter- node from tip), medium-hard stem of middle region (3rd or 4th internode from tip) and hard-woody stem of basal region (5th or 6th internode from tip) of one year old plant depending upon the growth phases of the stock plants. Grafting operations were performed as previously done in 1990 (Hossain et et., 1991 ) and with two types of incisions made on the stock plant (i) Superficial cut with very little xylem (wood) tissues and ( ii ) deep cut of approximately 1/4th of the diameter of stock internode. Scions of 'Lengra' variety and from the same seedling stocks of local types were used in different experiments. Following these studies, grafting on primary young branches on fall and short-sized plants of local types were done using 'Lenqra', 'Gopalbogh', 'Fazli" and a local type as scions. For micropropagation technique, experiments on decontamination of mango shoot tips were carried out using 1-5% NaOCl, 0.1% Hg Cl₂ and 6% H₂O₂. The sealing of cut ends of shoot tips, presterilisation and sterilisation followed by washing in the sterilised water were performed according to Hossain (1988). The shoot tips were then cultured in Murashige and Skoog (1962) medium at 25-28^oC for decontamination records before regeneration study couId be started.

Achievement/Findings:

The results indicate that mango shoot tips may be decontaminated with only 0.1% HgCl₂ to an acceptable level of contamination when treated for 5-10 minutes. To avoid oversterilisation, certain level of contamination may be accepted for plant materials (Hossain 1986, 1988). With these results of decontamination, subsequent experiments have been designed for hormone treatments for callus formation as well as for regeneration from mango shoot tips.
