Domestication and Cultivation of Agrocybe Aegerita on Straw and Sawdust Substrate

Abstract/ Executive Summary:

A new species of mushroom in Bangladesh, Agrocybe aegerita has been domesticated on sawdust and paddy straw substrate. Highest mycelium growth was observed on Malt extract agar media, at pH 7.0, temperature range 25 0 -27 ºC. Fruit body formation proceeded normally at 19-22 ºC and 3 times flashes were completed within 40 days, producing a total of 71.75 g on sawdust and 64.5 g on paddy straw at a biological efficiency of 32.1% and 28.3% respectively.

Objectives:

To find out the best cultivation media, pH and temperature range as well as carbon and nitrogen source for mycelial growth of Agrocybe aegerita.

Methodology:

The experiment was conducted in the tissue culture laboratory of National Mushroom Development and Extension Centre during July to October 2008. Agrocybe aegerita was collected from the National Mushroom Development and Extension center, Sobhanbagh, Savar, Dhaka. Fourteen different culture media Potato dextrose agar (PDA), Potato dextrose yeast agar (PDYA), Malt extract agar (MEA), Malt extract yeast agar (MEYA), Yeast extract agar (YEA), Wheat extract agar (WEA), Wheat extract yeast agar (WEYA), Cane juice agar (CJA), Cane juice yeast agar (CJYA), Corn extract agar (CEA), Corn extract yeast agar (CEYA), Sawdust extract agar (SEA), and Sawdust extract yeast agar (SYA) were used to investigate the mycelial growth of the mushroom. The media were adjusted to pH 6 and autoclaved for 20 minutes at 121 ºC and 1.5 kg/cm 2 pressure. Then 20 ml of each medium was aseptically poured into plates. After sterilization and solidification, the plates were inoculated with the inocula of Agrocybe aegerita mushroom. Average mycelial growth and duration of complete mycelium running of each replication was calculated. To determine a suitable temperature for the mycelial growth of Agrocybe aegerita, the medium (MEA) was adjusted to pH 6.0 and incubated at 5 different temperatures such as 15 ^oC, 20 ^oC, 25 ^oC, 30 ^oC and 35 ^oC. Average mycelial growth of each replication was calculated. The medium (MEA) was adjusted to pH 4.5, 5.0, 5.5, 6.0, 6.5, 7.0, 7.5, 8.0, 8.5 and 9.0 before autoclave and incubated at 25 ^oC. The measurement of mycelial growth was also performed following the same technique which was described earlier. S even media with different ratios of carbon (glucose) and nitrogen (Yest) such as (6:1, 12:1, 18:1, 24:1, 36:1, 48:1 and 96:1) were prepared to investigate the mycelial growth of Agrocybe aegerita. Then plates were transferred into incubation room for mycelium running at 25 ± 2 ^oC temperature. Average mycelial growth and duration of complete mycelium running of each replication was calculated. The experiment was laid out by following the completely randomized design (CRD) with 4 replicati ons. Data on mycelium growth rate (cm/day), duration of complete mycelium running (days), Number of Effective Fruiting body (NEFB), Days to Harvest (DH), Biological yield (BY), Economical Yield (EY) were recorded and analyzed following Gomez and Gomez, 1984 using MSTATC computer program. Means were computed following DMRT using the same computer program.
