The study was conducted on mycelial culture conditions, growth on differentt media and yield attributes and chemical compositions of Shaggy mushroom. This mushroom produced best mycelial growth (0.48cm/day) on malt extract agar medium followed by potato dextrose agar medium (0.44 cm/day) after incubation. The highest mycelial growth was observed at 23-25 oC temperature while no mycelial growth was observed at >35 oC or <10 oC. Likewise, very good growth was observed at pH 8.0 whereas the lowest growth was found at pH 5.0. Among the carbohydrate sources tested, maltose stimulated the highest growth and poor performance was exhibited in lactose. Carbon to nitrogen ratio of 12: 1 stimulated good for mycelial growth. In case of fruiting body production, saw dust, rice straw and mixed compost were used to identify the suitable substrate for Coprinus comatus. The highest biological yield was obtained in saw dust (146 g/packet) while no yield was obtained from mixed compost. The maximum carbohydrate and protein was found in fresh fruiting body of shaggy mushroom which was grown on saw dust but the highest fat, fiber and ash content observed in rice straw.

To find out the suitable agro-wastes substrates for reproductive growth.

This experiment was conducted in the tissue culture laboratory of National Mushroom Development and Extension Centre (NAMDEC) Savar, Dhaka during June to November, 2008. Pure culture of Coprinus comates was prepared on different media such as potato dextrose agar ( PDA) medium, malt extract agar medium (MEA), potato dextrose yeast agar (PDYA) medium, potato dextrose yeast agar (PDYA)medium and wheat extract agar medium(WEA). The basal component of each media was mixed with 20g of dextrose and 18g of agar at pH 6.5. The mixture was boiled on gas burner until the agar dissolved. The media was poured into Petri dishes (90mm diameter) at 20 ml/plates and sterilized in an autoclave for 20 minutes at 120 oC under 1 kg/cm 2 pressure. After sterilization and solidification, the plates were inoculated with the inocula of Coprinus comatus mushroom spices. Then plates were transferred in incubation room for mycelium running at 25oC temperature. Average mycelial growth and duration of complete mycelium running of each replication was calculated. Mother culture was prepared by mixing sawdust and wheat bran at the ratio of 2:1. Calcium carbonate was used at the rate of 0.2%of the mixture. The moisture level of the mixture was maintained at 65% by adding tap water. Polypropylene bags of 18x 25 cm size were filed with 200g of the above prepared mixture and packed tightly. The neck of the bag was plugged with cotton and covered with brown paper and tied with a rubber band. The packets were sterilized in an autoclaved for one hour at 120 oC less than 1kg/cm 2 pressure. After sterilization the packets were cooled for one hour and transferred into a clean bench. Three different substrates namely, sawdust, mature rice straw and mixed compost were tested in the experiment. The plant materials were cut into small pieces (0.5- 1 cm) and mixed preparing with nutrient materials, Polypropylene bags of 22.5 × 30 cm size were filled with 500g of substrate mixture. The procedure of packet preparation, plugging, sterilization and incubation were the same as mentioned under the section preparation of mother culture. Each spawn packet was inoculated with the mother culture at the rate of two teaspoonfuls per packet. After inoculation, the packets were incubated in the laboratory at about 27 oC temperature. Growth of mycelium in the spawn packets was completed within about 18 to 61 days. The experiment was laid out following completely randomized design (CRD) with four replications. Data on mycelium growth rate, duration of complete mycelium running, days to primordial initiation number, number of primordial initiation, number of effective fruiting body, days to harvest, biological yield, economical yield, length of stalk, diameter of stalk, length of pileus and diameter of pileus were recorded and analyzed following standard methods using MSTATC computer program. Means were computed following DMRT using the same computer program. The energy values of fruit bodies were calculated on the basis of their content of crude protein, carbohydrates, fat, fiber, ash and moisture.
