Studies on Immune Response of Chickens to Fowl Typhoid Vaccines

Abstract/ Executive Summary:

Chickens were immunized with experimentally prepared formalin-killed, heat-killed or live attenuated fowl typhoid vaccine to observe the immune response. Among the three vaccines, live attenuated vaccine and heat-killed vaccine induced almost equally higher antibody response than did the formalin-killed vaccine. Delayed type hypersensitivity test, however, did not produce any obviously positive reaction in any of the vaccinated groups

Objectives:

Immune response of chickens were compared after vaccination with a live attenuated vaccine and two killed FT vaccines prepared experimentally with a local strain of S. gallinarum

Methodology:

Chickens: White leghorn chickens of four months of age that had no previous history of vaccination against fowl typhoid were obtained from the Bangladesh Agricultural University poultry farm. The birds were observed for one month and the apparently healthy birds were selected for experimental use. Bacteria: The vaccine strain Salmonella gallinarum designated as SG5 was originally isolated from Mirpur Govt. Poultry Farm in collaboration with the Central Disease Investigation Laboratory (CDIL), Dhaka. The bacteria was found to be highly pathogenic and produced 100% mortality in 4 month old chicken after experimental infection (Begum, 1992). Vaccine: Three types of vaccines namely (a) formalin-killed (FK), (b) heat-killed (HK) and (c) live attenuated (LA) were prepared with the local isolate of S. gallinarum (SG5). FK vaccine was prepared following the procedure described for Pasteurella multocida by Choudhury et al (1987). HK vaccine was prepared by heat treatment of the cultured organism (60⁰C for 30 minutes in hot waterbath). The killed vaccines were stored in a refrigerator at 4-8⁰C. Live attenuated vaccine was prepared by serial passage of the bacteria in Nutrient agar media incubating alternately at high temperature (42⁰C) and at low temperature (25⁰C) upto 100 times. The loss of virulence was tested by subcutaneous inoculation of the culture into 10 white mice at the rate of 0.2 ml per animal. None of the mice died or showed any apparent sign of infection during the period or 14 days of observation. This culture was stored in a refrigerator for future use. Bacteriological sterility, purity and safety tests of the experimental vaccines, as appropriate, were conducted as per the method described by Heddleston and Reisinger (1960) and Matsumoto and Helfer (1977). Vaccination: For vaccination a total of 60 chickens of either sex of four months or age were used. The birds were randomly divided into three equal groups. Each group was vaccinated twice at 2 weeks interval with one of the three vaccines given by the intramuscular route at the rate of I ml per chicken.

Achievement/Findings:

The present study indicated that both LA and HK vaccines, but not the FK vaccine, induced good humoral immune response. However, the relative efficacy of LA and HK vaccines could not be ascertained as a protection trial was not conducted. Further studies in this regard are now in progress.
