The present investigation was carried out to find out the effect of different bed size and ages of media on the growth, yield and quality of straw mushroom. Pasteurized rice straw used as substrate. The seven different bed sizes such as T1 = (15 cm × 15 cm× 1m), T2 = (20 cm × 20 cm× 1m), T3 = (25 cm × 25 cm× 1m), T4 = (30 cm × 30 cm× 1m), T5 = (35 cm × 35 cm× 1m), T6 = (40 cm × 40 cm× 1m), T7 = (45 cm × 45 cm× 1m) and various incubation intervals (10, 15, 20, 25, 30 and 40 days) were used as treatments. In case of bed sizes, the maximum yield of mushroom was obtained from T7 (1497.00 g/bed) followed by the T6, T5 and T4 T1 gave the lowest yield (159.00 g/bed). In case of ages of media, the yield increased gradually up to 20 days and decreased gradually thereafter. Its yield was 738.80, 912.30, 1029.00, 776.50, 601.50 and 421.00 g/bed at the ages of 10, 15, 20, 25, 30 and 40 days respectively. On the basis of the present findings, it may be explained that it is possible to obtain significantly higher yields of V. volvacea by cultivating in T7 = (45 cm × 45 cm× 1m) and the highest biological efficiency was found in T4 (23.38%), beds using 15 and 20 days old spawn with well developed mycelium.

To find out the effect of different bed sizes for the yield and to determine the ages of media for biological efficiency of straw mushroom.

There are several media on which the mushroom culture can grow, such as potato dextrose agar (PDA), malt extract agar (MEA), potato yeast dextrose agar (PDYA) and Yeast extract agar (YA). The compositions of potato dextrose agar (PDA) are given below: Washing, peeling and slicing of 200g potatoes. Boiling in 1000 ml distilled water until potatoes become soft enough to be eaten but not over cooked. Straining through cheese cloth & collecting of liquid in graduated cylinder followed by restoring of volume to 1000ml by adding fresh distilled water. The basal component of the media was mixed 20 g of dextrose and 20 g of agar at pH 6.5. The media was boiled on gas burner until the agar dissolved. The media was poured into Petri dishes (90 mm diameter) at 20 ml/plates and sterilized in an autoclave for 20 minutes at 1200C under 1kg/cm2 pressure. After sterilization and solidification, the plates were transferred in to incubation room for mycelium running at 250C temperature. The protocols of rice straw substrates are mentioned below: At first the rice straw cut into pieces of 2.5 to 5 cm long. Then rice straw was soaked in water for 2 to 4 hours. Wheat barn was mixed @ 30% wet basis and the polypropylene bags of 7'' × 10'' size were filled with 300 g of substrate and their mouths were plugged by inserting water absorbing cotton with the help of plastic neck. The bags were autoclaved at 1210C and 1.5 kg/cm2 pressure for 2 hour. After autoclaving and cooling, the bags were inoculated with the tissue culture of Volvariella volvacea. Then, the inoculated packets were kept for mycelium running at 250-300C. The substrate was pasteurized using steam for 60 minutes at 600C in a drum. The pasteurized straw was spread on a clean cement floor inside room and allowed to cool to room temperature for 12 hours. The different treatments for bed sizes as follow: T1 = (15 cm × 15 cm × 1 m), T2 = (20 cm × 20 cm× 1 m), T3 =(25 cm × 25 cm × 1 m) ,T4= (30 cm × 30 cm × 1 m),T5= (35 cm × 35 cm × 1 m), T6= 40 cm × 40 cm × 1 m), T7 =(45 cm × 45 cm× 1 m). For the experiment, seven treatments were used. Before spawning, 65% moisture was maintained with the substrate. A wooden rack of different size mention above was used for the bed preparation. At first straw substrate sprayed in the wooden rack with 10 cm thickness and mother culture mixed thoroughly on the surface. Observations were recorded with respect to Days for mycelium run (DFMR), days for primordial initiation (DFPI), days for first harvest (DFH), fruiting body/bed (number), average weight/fruiting(g) and yield parameters as described earlier. Biological efficiency was determined by the following formula: Biological Efficiency (%) = {Total biological yield (g) / Total substrate used (g) } x 100.
