Effects of Different Temperature, Humidity and Ph on Mycelliall Growth of Beauveria Assiana, the Pathogen of White Muscardine Disease of Silkworm Bombyx Mori L.

Abstract/ Executive Summary:

Effects of different temperatures, humidities and pH values on mycellial growth of Beauveria bassana, the pathogen of white muscardine disease of silkworm Bombyx mori L were studied. The maximum mycellial growth was recorded at 25°C, 100 percent relative humidity and pH value of 5 after nine days of incubation. The results reveal that temperature, humidity and pH have profound effect on mycellial growth.

Objectives:

To find out the actual influence of the different factors on mycellial growth in culture medium.

Methodology:

Peptone dextrose agar, IN Hcl, IN NaoH, petriplates, desiccator and large size incubator were taken as materials for this experiment. Beauveria bassiana (Bals) Yuill the organism was isolated from white muscardine diseased worms and cultured on peptone dextrose agar medium. Ten days old culture of the fungus was used for the experiment. Seven temperatures such as 5, 10, 15, 20, 25, 30, and 35°C were selected and adjusted in incubator for the test of fungal growth. Six relative humidities 74, 86, 90, 95, 97, and 100 percent were adjusted by using glycerol and distilled water in desiccators. On the other hand arrangement of nine pH such as 2, 3, 4, 5, 6, 7, 8, 9, and 10 were prepared by using IN HCl and NaOH. In every case, 2mm culture block of B. bassiana was cut from the periphery of the ten days old culture and placed at the center of the peptone dextrose agar containing 90 mm petriplates. The petriplates with three replication were then placed in desiccators and finally in Incubator up to nine days for every experiments. The diameter of the fungal colony either circular or irregular was measured.

Achievement/Findings:

It is evident from the experiment that the fungus prefers acidic media for its growth than the basic media. The above findings indicate that the temperature, humidity and pH have profound effect on growth of fungus in culture.
