Distribution of Free Amino Acids and Sugars in Tea Soils of Bangladesh

Abstract/ Executive Summary:

Amino acids identified were serine, aspertic acid, alanine, valine, methionine, iso-leucine, leucine and histidine, Glucose, galactose arabinose and xylose were found from all the 9 soil samples investigated for sugars. No relationship was obtained between the occurance of amino acids and productivity of the soils. A relationship was however obtained between the quantity of amino acids and the productivity of the soils. High yielding soils contained less amount of amino acid. Topography had little effect on the distribution of amino acids.

Objectives:

It was partitioning chromatography made the detection of small quantities of amino acids and sugars possible, and was adapted in the present investigation.

Methodology:

Soil samples representing 0-9 inches and differing in productivity were collected from Tea Research Station, Sreemangal in the District of Sylhet, The samples were kept inside polythene bags, sealed and were stored in refrigerator until analysis were made.sodium hydroxide (d) IN sodium chloride (e) 6N HCI and 6N H₂SO₄ were tried for extraction of amino acids and sugars .The first four extracting agents, a, b, c & d failed to extract either amino acid or sugar. Extractants e and f some spots but they were either continuous or indistinct. Hot water extract tried as a new method produced highly satisfactory result and was used for extracting both amino acids and sugars. The solution ratio was 1 : 4 (50 gms of soil and 200 ml of distilled water) and time of extraction was 10 hours under reflux condition. The extract to be analysed were applied to the filter paper sheets in drops of 0.18 ml. to 0.24 ml by means of a 60 micro pipette for the identification of both amino acids and sugars. The drops were spaced at intervals of 1 inch to l1/2 inches in a line, l1/2 inches from foot of the paper sheets and not closer than 1 inch to the edges as described by Doreen et al, 1950. A portions of the solvents butanol : acetic acid : water for amino acid and butanol : pyridine : water for sugars, were placed in at the bottom of two different cylinders. Filter paper sheets with the spots were first hanged inside the cylinders for 5 to 6 hours for saturation in presence of suitable solvent. The sheets were then fastened into cylinder so that the edges were barely touching and the sides along which the spots were applied were immersed in the solvent kept in a glass boat placed at the top of the cylinder. The solvent was allowed to descend at a depth of approximately 35 cm. the time required to descend varied from 12 to 16 hours for amino acids and 10 to 14 hours for sugars. The paper sheet were then removed from the cylinder and allowed to dry over night. The nynhydrin solution was sprayed on for the detection of amino acids and a solution of aniline, pthalic and glacial acetic acid for sugar. After the spray, the sheets were dried and put into an oven at 80ºC for 5 minutes when spots of different amino acids and sugars appeared. The amino acids were then indentified by their Rf. values given by Block et al. The sugars were identified by comparing the distances of individual sugars with those of known sugars. Quantitative estimation of amino acids were done by partition of the amino acids and development of the spots were carried out as in the qualitative method except that 240 ml of the extract was used for giving the spots. Each spot with a 1/8 inch margin around it was cut out of the paper and placed in a cleaned dry test tube. The colour was extracted with l0 ml of acetone and allowed to stand about one hour before being read on a colorimeter with a 570 mu filter. these were then compared with known quantities of standard amino acids.
