Effecty of Water Logging on Growth Parameters and Stomatal and Leaf Anatomical Characters of Mulberry

Abstract/ Executive Summary:

Experiment was conducted to study the effect of waterlogging on top dry weight, leaf area growth parameters and stomatal and leaf anatomical characters of five mulberry varieties. There were five treatments. Both leaf dry weight and leaf area were highest in the normal watering treatment compared to the waterlogging treatment. Specific leaf area (SLA) was better in normal watering and leaf weight ratio (LWR) in the waterlogged treatments. Normal watering had the highest values for stomatal frequency, guard cell length, leaf thickness and cuticle thickness. Among the waterlogging treatments, T₄ plants (at 60 days after planting, the pots were dipped for 15 days) had less and T₃ (at 30 DAP, pots were dipped for 30 days) had less severe effect for stomatal characters.

Objectives:

Tostudy the effect of waterlogging on growth and stomatal and leaf anatomical characters of five mulberry varieties.

Methodology:

Five improved mulberry (Morns alba L.) varieties used were BM-I, BM-2, BM-3, BM-4 and BM-5. The experiment was carried out in large earthen pots of 35cm height and 50cm diameter at the top and 20cm at the bottom. The pots were arranged in a randomized block design with three replications. Five plants were maintained in each pot. There were five treatments: T₁- Normal watering as and when necessary, T₂ - at 30 days after planting (DAP) the pots dipped in water for 15 days, T₃ - at 30 DAP the pots were dipped in water for 30 days, T₄ - at 60 DAP the pots were dipped in water for 15 days and T 5-at 60 DAP the pots were dipped in water for 30 days. Plants were waterlogging by submerging the pots in earthen bowls and water level was always kept 2-3cm above the soil surface of the pots. The experiment was conducted in two seasons in a year, i.e., February-April and November-January, There were four harvests in each season. Measurements of dry weights of stem and leaves and leaf area were taken and SLA and LWR were calculated. Fifth to seventh leaves from ninety days old shoots were collected at 10am in a beaker containing water. Subsequently a thin film of Quick fix was applied on the central portion of the lower surface of the leaf blade. Stomatal impressions were taken and the frequency was determined. Twenty fields were recorded and the frequency of stomata per mm² was estimated. The average length and breadth of the guard cell was deteinlined on twenty measurements. For the measurement of leaf and cuticle thickness, hand sections were taken from three stages of leaves (tender, medium and mature) and the sections were stained with 1% safranine and mounted in 5% glycerin. Leaf thickness and cuticle thickness were measured in micrometer. Ten readings were taken and the mean was calculated for final recording. Data were analysed statistically.
