A Study on the Comparison of Three Incubation Methods for Evaluating Mineralization of Organic Phosphorus in Soils

Abstract/ Executive Summary:

An experiment was carried out to compare the effect of three incubation methods upon the mineralization of organic phosphorus in soils over a period of 60 days. Mineralization was greater in the barium peroxide method than in the other two methods included in the investigation.

Objectives:

To give a comparative view of organic phosphorus mineralized in three different incubation methods

Methodology:

10 g samples of air dried 2 mm sieved clay loam (Jaflong series - pH 4. 1) and clay (Lauta series - pH 4.7) were weighed into test tubes (16 cm x 2 cm). Water was added to bring the moisture content to 50 per cent of the water holding capacity. Three types of incubation procedures were used. 1) Open tube method - Soils were incubated in test tubes with mouths open throughout the incubation period. Soil moisture was kept constant during the experiment by the daily addition of the required amount of distilled water. 2) Occasional aeration method - Soils were sealed in test tubes but aerated every 72 hours and enough water was added to make up for any moisture lost (Acquaye¹). 3) Barium peroxide method - In the closed test tubes vials containing barium peroxide were inserted to supply oxygen and absorb carbon dioxide during incubation (Cornfield²). A temperature of 25⁰c was maintained throughout the incubation experiment. Soils were analyzed-for mineralized organic phosphorus on 0, 7, 15, 30, 45 and 60 days from the day of incubation. The difference between the 0.2 N H₂SO₄ extractable phosphorus before and after incubation was taken as the measure of the amount of organic phosphorus mineralized.

Achievement/Findings:

The amount of organic phosphorus mineralized during 60 days of incubation period and the amount of organic phosphorus mineralized usually increased with period of incubation .values of organic phosphorus mineralized varied significantly from method to method except those measured after 7 and 30 days with the Lauta soil. The barium peroxide method yielded the highest amount of phosphorus mineralized followed by the occasional aeration method while the open tube method accounted for the least value. The barium peroxide ensuring an uniform and constant supply of oxygen with carbon dioxide absorption was most effective in producing a condition conducive to the mineralization of organic phosphorus in soils. Therefore, it is suggested that the barium peroxide method be used for studying mineralization of organic phosphorus in soils.
