Five native bacterial strains viz. Bacillus sp. SMH/2, Bacillus sp. GLL/5, Cellulomonas sp SWTQ/3, Micrococcus sp. dump T/1 and Pseudomonas sp SS/2 were artificially inoculated for rapid degradation of organic solid waste. Micrococcus sp. dump T/1 was found the most effective strain and showed the highest weight loss 27.65% when spore suspension was used, and it was 12.67% greater than the control. Spore suspension was found more effective than the culture disc treatment, and autoclaved OSW was better than the non autoclaved OSW for degradation. Degradation of OSW was also found to increase with the increase in concentration of molasses but decrease with the increase in concentration of sugar and significant (P = 0.05) percentage of weight loss was observed at different concentrations. The highest weight loss was observed in 15 % molasses, which was 20% greater than control and 9% more than without additives when bacterial suspension was used.

  2. To find out the better additives for adaptation of bacteria that may enhance the degradation process of organic solid waste.

For effective decomposition of organic solid waste five native bacterial strains such as Bacillus sp. SMH/2, Bacillus sp. GLL/5, Cellulomonas sp. SWTQ/3, Micrococcus sp. dump T/1 and Pseudomonas sp. SS/2 were collected from stock culture of Biotechnology and Microbiology Laboratory, Department of Botany, Rajshahi University which were previously isolated from soil, humus and garbage samples (Rahman, 2008). Organic solid waste (OSW) was collected from different garbage centers of Rajshahi University campus using polythene bags and was kept under room temperature until use. Aseptic condition was maintained as far as possible during collection. The kitchen wastes were cut into 2-3 mm size and weighed into 100g samples, and kept in per 150 ml bottle with cotton plug. Both autoclaved and no autoclaved kitchen waste (including bottle) were used. Bacillus sp. SMH/2, Bacillus sp. were cultured on NA medium and GLL/5, Cellulomonas sp. SWTQ/3, Micrococcus sp. dump T/1 and Pseudomonas sp.SS/2 were cultured on BCDA medium at 30 °C for 24 hours. Then culture discs (5 mm diam.) were cut with cork borer and added to autoclaved and non autoclaved kitchen waste separately at the rate of 5 discs/100g sample. All the steps were completed inside the running laminar airflow. Control treatments were performed with no inoculation. Sterile thermometer was pushed through cotton plug for each bottle. Then the bottles were sealed with Para film and labeled and inoculated for 30 days at 28 ± 2 °C. Ten ml of autoclaved distilled water was added 24 hours old bacterial culture tube and mixed well. After that 10 ml of spore solution was added to 100 gm autoclaved and non autoclaved kitchen wastes separately and mixed well. Control treatments were performed with no inoculation. As well as culture disc experiment sterile thermometer was used and the bottles were sealed with Para film and incubated for 30 days at 28± 2 °C.

From the above findings it was noted that Micrococcus sp. dump T/1 was the most potential strain for bioconversion of organic solid waste which was more active on autoclaved waste than non autoclaved organic solid waste. Bacterial suspension was more active than culture disc treatment. Molasses was the better additive than sucrose to enhance the decomposition process. So, it may be concluded that Micrococcus sp. dump T/1 may be used in preparing low price inputs (compost) within short period and molasses can be used as additives to enhance of biodegradation process.
