Establishment of High Frequency Shoot Regeneration Method from Cotyledon Explants in Jute (corchorus Capsularis L.)

Abstract/ Executive Summary:

An efficient tissue culture method for a high rate of shoot regeneration was developed for Corchorus capsularis L. var. CVE-3 (Jute). Cotyledonary explants with petiole were cultured in MS medium supplemented with auxin and cytokinin. The best shoot proliferation was observed when cotyledon derived calli was subcultured in medium fortified with a combination of 2.0 mg /1 BAP and 0.5 mg/L IAA. Elongated shoots were rooted with a 95% success by treating them with MSO (MS medium without hormone) medium. Rooted plants were successfully established into soil and the survival rate of the plants was more than 80%. The regenerant matured as normal and produced fertile seeds.

Objectives:

To study on establishment of high frequency shoot regeneration method from cotyledon explants in jute (Corchorus capsularis L.)

Methodology:

Germination of seeds: Seeds of Corchorus capsularis, variety (CVE-3) and C.olitorius (O-72) were surface sterilized by immersing in 0.1% (w/v) mercuric chloride for 20 min, followed by 6 washes with sterile deionized water. Seeds were placed on the surface of 50 ml aliquots of hormone free agar-solidified (0.8%, w/v) MS basal medium (Murashige and Skoog, 1962) contained in 100 ml capacity conical flasks. In another set of experiment, surgical cotton (1 g approx, in each flask) was used instead of agar in association with MS basal liquid medium to obtain optimum seedling production. Surgical cotton was placed at the bottom of 100 ml flasks. Each flask contained 20 ml of hormone free MS liquid medium. Cultures were placed in a growth room with 28ºC under 1.0 Wm^-2 of daylight fluorescent tubes with 12 h photoperiod. Culture of cotyledons with attached petioles: The cotyledons (with attached petioles) of 7 day old in vitro grown seedlings of Corchorus capsularis (CVE-3) were excised and cultured on MS agar (0.8% w/v) solidified medium with 3% (w/v) sucrose and pH 5.8 with the supplement of IAA 0.5 mg t^-1 and with BAP (2.0 mg l^-1). Explants were cultured in 250 ml conical flasks and maintained at 28ºC under a 1.0 Wm^-2 of daylight fluorescent tubes with 12 h day length. The experiments were repeated 3 times with 3 replications. In three replication containing explants were placed under fluorescent light in a growth room with controlled temperature (28⁰C). The flasks were checked daily to note the appearance of callus and shoot regeneration. The results were scored six weeks after culture.

Achievement/Findings:

The ultimate goal of in vitro technique is production of free-living plantlets via shoot and root formation from callus. High responsive genotype CVE-3 for callus induction had high regeneration capacity, indicating that callus induction capacity is related to regeneration of shoot. This result correlated with the findings of Khatun (2001) who reported that CVE-3 had high shoot regeneration capacity. Shoot regeneration from cultured cotyledons were transferred to MSO (MS medium without hormone) medium in order to induce root from the differentiated shoots. Rooted plantlets were transferred to pots after a short period of indoor acclimatization. The plants were vigorous and uniform and grew into maturity in four months after planting. Pods in mature plants were normal with well developed seeds.
