IRBB60 was used as donors of Bacterial Blight (BB) resistance genes, Xa21 and xa13 for pyramiding the Bacterial Blight (BB) resistance genes in BRRI dhan29. After molecular confirmation, BB screening was carried out on BC5F1 progenies and 3 resistant lines were selected. In Boro13-14, 3 selected lines were evaluated as observational trial . Selected plants were reconfirmed by further molecular analysis as well as bacterial inoculation with BXo9 isolate and finally two BB pyramided lines were selected .

To pyramid two BB resistance genes (Xa413 & Xa21) in BRRI dhan29

.

In this study, IRBB60 was used as donor plant and BRRI dhan29 were used as a recurrent parent. Thirty five days old single seedlings were transplanted in a 3.0 m × 2 row plot with 25 × 15 cm spacing. Fertilizers dose was 80:60:40 kg/ha NPK. Total amount of P and K fertilizers were applied at the time of final land preparation. Nitrogen fertilizer was applied in three splits @ (40+20+20) at 15, 30, 50 days after transplanting. After flowering, emasculation was done on BRRI dhan29 and then pollinated with  pollen from IRBB60 plant. to create F1 plant.. Several times backcrossing were done with BRRI dhan29 to create BC5F1 progenies. In each backcrossing, molecular characterization was carried out to ensure the presence of Bacterial Blight resistant gene ( Xa21 & xa13) in the progenies. Bacterial inoculation was done in BC5F1 plants during booting stage using clipping method and 14 days after bacterial inoculation, BB scoring was taken.
