Screening for Cold Tolerance of BRRI Gene Bank Germplasm at Seedling Stage Under Natural Condition During Boro

Abstract/ Executive Summary:

A study was carried out through Yoshidas’ culture solution to find out ?rice germplasm tolerant to cold at seedling stage at Bangladesh Rice Research Institute. A severe cold spell (bellow 10 ⁰C were occurred from 8 to 13 January 2013 after sowing of seed resulting whitish or albino of all seedlings up to 23 days and then seedlings of some genotypes recovered and became green with the rising of temperature. The mean score was considered for the selection of a particular genotype. Rice genotypes having a mean score of <5 were considered as tolerant to low temperature at seedling stage. Among 87 genotypes, only 10 genotypes ( showed visual score 3 to 5 that is tolerant to moderately tolerant . Above all, ?BRRI Acc no-114, BRRI Acc no no-177, ?BRRI Acc no-197 and BRRI Acc no-202 were the most tolerant those showed mean score of 3. However, other genotypes showed visual score 7 to 9 that is susceptible to highly susceptible.

Objectives:

To identify rice genotypes which can tolerate low temperature at seedling stage.

Methodology:

Sprouted seeds of 87 rice germplasm collected from BRRI gene bank along with 3 check varieties namely BR18, BRRI dhan28 and BRRI dhan36 were sown on December 27, 2012 in netted bed attached with styro-foam sheet floating in a tray containing full strength culture solution. The pH level of culture solution was maintained at 5.0 throughout the experimental period. Temperature of the culture solution was taken at 9.00 am, 2.00 pm and 5.00 pm by a thermometer. Each entry was sown in a line and 10 seedlings in each line were maintained. Seedlings were grown up to 40 days and scored following the SES for cold tolerance.

Among 87 tested genotypes 10 genotypes (BRRI Acc no. 79, ?BRRIAcc no.102, ?BRRIAcc no.113, ?BRRIAcc no.114,BRRI Acc no.121, ?BRRIAcc no.127, ?BRRIAcc no.169, ?BRRIAcc no.177, ?BRRI Acc no.197 and ?BRRIAcc no.202) were selected for further studies.
