Result of the experiment showed that most of the tested isolates were effective to inhibit mycelial growth of S. rolfsii and R. solani on culture media. The isolates RT1, 2M2, GT11, 5M1 and RT46 caused highest reduction (78 to 81%) of mycelial growth of S. rolfsii and the isolates, RT1 showed the highest growth inhibition. On the other hand the isolates GT1, TS9, 3M2, 2M1 and RT1 caused highest reduction (74 to 82%) of mycelial growth of R. solani.  GT1 isolates of T. harzianum showed highest inhibition of both of S. rolfsii (80%) and R. solani (82%).

       The present research works were undertaken to screen the potential isolates of Trichoderma spp. against Sclerotium rolfsii and Rhizoctonia solani

The experiment was conducted at the laboratory of Plant Pathology Division, BARI, Gazaipur during 2004-05. Trichoderma harzianum isolates were collected and isolated from the rhizosphere and rhizoplane of different crops of different agroecological zone in Bangladesh by soil dilution plate and root washing method. An in-vitro study was conducted to find out the antagonistic effect of 40 selected T. harzianum isolates against S. rolfsii and R. solani on PDA by dual culture technique. Three discs of each isolate were placed at the edges of each PDA plate and one disc of S. rolfsii was placed in the centre. The plates only with the discs of two test pathogens in the center were used as control plate. The plates were then incubated at room temperature (29 ±1°C) until the mycelium of test pathogens covered the whole plate. The plates were arranged in Completely Randomized Design (CRD) with three replications. The diameter of the colonies was measured after the complete growth of the control plates. There after inhibition percentages of the test pathogens were calculated based on the growth of the pathogen on PDA plates in absence of antagonistic fungal isolates following the formula as suggested by Sundar et al. (1995).

Isolates were classified on the basis of inhibiting potentiality of incubation. The fungal isolates which inhibited around 75-90% absolute inhibition of the two test pathogens were grouped as very strong, 60-75% inhibition were grouped as strong, 50-60% inhibition were grouped as moderate, 30-50% inhibition were grouped as weak, 15-30% inhibition were grouped as very weak, and 0-15% inhibition were grouped as no inhibition (Sundar et al. 1995). Isolates having strong suppressive effect on mycelial growth of the two test pathogens were selected for further study.

Forty effective isolates of T. harzianum were tested against S. rolfsii and R. solani isolates by dual plate culture technique on PDA. All the test isolates reduced the growth of S. rolfsii and R. solani as compared to control. Among the isolates, RT1 showed the highest growth inhibition (81%) of S. rolfsii while GT7 showed minimum growth reduction of S. rolfsii (60%). The isolates RT1, 2M2, GT11, 5M1 and RT46 caused highest reduction (78 to 81%) of mycelial growth of S. rolfsii while GT7, WRT44, 2M1 and TW108 gave the lowest reduction of 60 to 62% inhibition of radial mycelial growth of S. rolfsii.  5M1 showed the highest growth inhibition (82%) of R. solani while T4 showed minimum growth reduction. The isolates GT1, TS9, 3M2, 2M1 and RT1 caused highest reduction (74 to 82%) of mycelial growth of R. solani while T4, TW108, T5 and GT5 gave the lowest reduction of 33 to 50% inhibition. GT1 isolates of T. harzianum showed highest inhibition of both of S. rolfsii (80%) and R. solani (82%).
