In Vitro Study on Some Isolates of Trichoderma Spp for Their Compatibility with Fungicides.

Abstract/ Executive Summary:

A study was under taken during 2004-05 at the laboratory of Plant Pathology Division, BARI, Gazipur to test the compatibility of Trichoderma spp. with some fungicides in-vitro. The test fungicides were Vitavax-200, PCNB, Bavistin and Rovral at 100, 250, 500 and 1000 ppm. The results indicated that the isolate (GT1) of Trichoderma spp. was highly sensitive to Bavistin at all concentration. The isolate (GT1) was found incompatible with the Rovral, Vitavax-200 and PCNB at lower doses (100 and 250 ppm) only.

Objectives:

A study was under taken to test the compatibility of Trichoderma spp. with some fungicides. Four fungicides namely, Vitavax-200, PCNB, Bavistin and Rovral at 100, 250, 500 and 1000 ppm concentrations were tested by Poison food technique.

Methodology:

An in-vitro experiment was conducted to find out the compatibility of T. harzianum isolates with four selected fungicides namely Vitavax-200, PCNB, Bavistin and Rovral at 100, 250, 500 and 1000 ppm concentrations by Poison food technique (Dhingra and Sinclair 1985 and Begum 2000). Requisite amount of fungicide was added to 100 ml PDA in 100 ml conical flask before sterilization. The PDA plates without fungicides were used as control. The percent inhibition of the radial growth was recorded fourth days after incubation when the control plates were covered with the full growth of Trichoderma. Inhibition of radial growth was computed based on colony diameter on control plate using the formula of Sundar et al. (1995) as already described.

Achievement/Findings:

Compatibility of T. harzianum isolates, GT1 with selected fungicides: The effect of fungicides on radial mycelial diameter of Trichoderma isolate GT1 were tested on PDA and the results are shown in Table 3. Among the fungicides, Bavistin 50WP at 100, 250, 500 and 1000 ppm completely inhibited the growth of Trichoderma isolate (GT1). It indicated that the isolate of the antagonist is highly sensitive to Bavistin and it is not compatible with the fungicide. The inhibition of colony diameter of the Trichoderma isolate at 100, 250, 500 and 1000 ppm of Rovral 50WP was 22.04, 30.94, 41.11 and 53.52%, respectively. It revealed that the isolate GT1 was less sesitive Rovral at lower doses only. At 500 and 1000 ppm of Vitavax-200, inhibition of colony growth was 76.00 and 88.00%. At 100 and 250 ppm of Vitavax-200, colony growth was inhibited by 5.55 and 45.90%, respectively. Complete inhibition of colony growth of Trichoderma harzianum isolate was observed at only 1000 ppm of PCNB. At three lower concentrations PCNB caused 14.44 - 31.66% growth inhibition of the Trichoderma spp. It indicated that GT1 was highly compatible with the lowest concentration of Vitavax and PCNB. Trichoderma harzianum was found highly incompatible at all the concentrations of Bavistin, two higher concentrations of Vitavax-200 and the highest concentration of PCNB.
