Microbiological Quality of Different Companies Mineral and Drinking Water Available in Dhaka City

Abstract/ Executive Summary:

Bottled mineral water and drinking water of ten companies were collected from different departmental stores of Dhaka city. Results indicated that both mineral and drinking water were contaminated by bacteria, fungus and worm. In case of mineral water bacterial counts varied between OAxlO' to 5xl03 cfu ml", where as in drinking water the range was 0.3xlO' to 8xl03 cfu ml". Fungus was present in mineral water, except one, though the number was low. Worm was also present in both mineral and drinking water.

Objectives:

1. To assess quality of bottled mineral water and drinking water of ten companies.

Methodology:

Bottled mineral water and drinking water of ten companies were randomly collected from different departmental stores of Dhaka city. Standard methods were followed for all bacteriological techniques and the time and temperature of incubation. Total viable counts of bacteria and fungus were done by using nutrient agar and potato dextrose agar media respectively. In the standard multiple tube fermentation technique for coliform detection, different amounts of water (viz, 10.1, and 0.1 ml) samples were incubated at 37° C for 48h. The production of gas in the tube, which is indicative of positive presumptive test, were enumerated through consu-ltation of MPN (Most Probable Number) table. All the tubes which showed positive presumptive test after 48h were further subjected to confirmatory test. The bacteria were inoculated in confirmatory test medium which was a brilliant green lactose bile broth, and were incubated at 37°C for 48h. Gas production indicated positive confirmed test. Bacteria from the tubes of presumptive positive test were streaked on eosin ethylene blue medium. Typical and atypical colonies were picked up from EMB agar medium and the organisms were streaked on a sland of nutrient agar. After 24h of incubation at 37°C a small portion of growth from the slant was used for gram staining and another portion was transferred into secondary broth tubes of large tryptose broth. Non spore forming gram negative rod shaped bacteria with gas formation positive in the second tryptose broth tube indicated that completed test was satisfactori ly positive.
