Screening for Salinity Tolerance of Some Anther Culture Lines of Rice at the Seedling Stage

Abstract/ Executive Summary:

The experiment was conducted at to screen out salt tolerant lines at seedling stage. Fifteen anther culture lines were screened at high saline condition using hydroponic system. Among 15 lines, only 3 lines as Bio-AC1B-19-2-1-2-4, Bio-AC1B-19-2-1-3-2, Bio-AC1B-19-2-1-5-2 showed visual score 5 that is moderately tolerant. However, other genotypes shows visual score 6 to 9 that is susceptible to highly susceptible at high saline condition.The survival percentage of these lines ranged 60 to 70%.

Objectives:

To screen out some anther culture lines against salinity tolerance at the seedling stage.

Methodology:

Fifteen anther culture lines along with tolerant and susceptible check were considered for this experiment. Sprouted seeds were sown on Nylon net fitted with Styro-foam floating on Yoshida full strength culture solution according to Gregorio et al., 1997. Salinity stress @ 12 dS/m was applied 7 days after sowing. The culture solution was changed once a week throughout the experimental period. The pH and EC of the culture solution was checked daily and maintained at 5.0 and 12 dS/m respectively. In each tray FL478 and IR29 were used as a tolerant and susceptible check, respectively. When susceptible check IR29 died, visual score was made according to SES system and living plants were recorded. The experiment was conducted in RCB design with 2 replications.

Achievement/Findings:

Three (3) genotypes were identified as moderately tolerant to salt stress at the seedling stage. These genotypes could be used for future breeding program to develop salt tolerant variety.
