Micropropagation of Averrhoa Carambola Linn. (cararnbola) by Tissue Culture

Abstract/ Executive Summary:

Auerrhoa carambola Linn. was micropropagated from seedling explants through forced axillary branching on Murashige and Skoog's (MS) and Linsmaier and Skoog's (LS) media supplemented with benzyladenine (BA) or kinetin. High frequency shoot formation was achieved on MS medium supplemented with 0.5 mg/l BA. Shoots could be rooted by treating them with indolebutyric acid (IBA) in half-strength MS medium. Regenerated plantlets were successfully established in soil after a short period of indoor acclimatization.

Objectives:

To reports the optimal culture conditions for axillary shoot proliferation from seedling nodal explants of A. carambola.

Methodology:

We collected seeds from unripe mature green carambola fruits (sweet type) and washed them thoroughly under running tap water. The material was surface sterilized with HgC1₂ (0.1 % w/v) for 10 minutes containing few drops of Tween-80 followed by 3-4 rinses in sterile distilled water. The decoated seeds were germinated on MS medium supplemented with 1mg/1BA. Then we excised single node stem segments from 30-day-old seedlings and cultured them on MS and LS basal media variously supplemented with BA and kinetin and transferred the regenerated shoots to half-strength MS medium with 0.1 to 2.0 mg/1 IBA for adventitious rooting. The pH of the medium was adjusted to 5.8±0.1 before addition of agar. After melting with 0.7% agar (BDH), the media were dispensed into culture vessels. All media were autoclaved for 20 minutes at 121°C (108 KPa). Cultures were maintained at 26±2°C with a 16h photoperiod. Ten to twelve replications were tested each treatment and experiments were repeated at least twice.
