Effect of Naci and Na2so4 on Germination, Callus Induction and Subsequent Plant Regeneration in Some BRRI Developed Rice Varieties

Abstract/ Executive Summary:

The experiment was conducted to evaluate the effects of NaCI and Na₂SO₄ on germination, callus induction and subsequent plant regeneration of 14 BRRI developed rice varieties at the Biotechnology laboratory, BRRI in 2007. Sterile dehusked rice seeds were placed on MS media supplemented with NaCI and Na₂SO₄at six concentration, (0. 1.0,2.0. 3.0,4.0 and 5.0 g/L). After germination and callus induction, the embryogenic calli were transferred to regeneration media. All the test parameters were affected sorely by Na₂SO₄ followed by NaCI. The parameters were highly impinged at 5.0 g/L salt concentration compared to control (0 g/L salt). Out of 14 test varieties, BRRI dhan 41 performed best for germination (9846%) and callus induction (95.31%), but BRRI dhan38 was good for plant regeneration (87.50%). Germination was not badly affected due to addition of salts in the callus induction media. However, regeneration was found highly affected from calli developed on the media containing salts. BRRI dhan41 was found as a component variety for germination, callus induction at lower salt loading while BRRI dhan38 was identify as a potential material for regeneration at lower salt level. So, the variety can be used as a potential material for transformation.

Objectives:

To evaluate the effect of NaCI and N₂,S0₄ on germination, callus induction and regeneration of 14 BRRI developed rice varieties.

Methodology:

Mature dehusked seeds of 14 BRRI developed rice varieties (BRRI dhan32, BRRI dhan33, BRRI dhan34, BRRI dhan35, BRRI dhan36, BRR! dhan37, BRRI dhan38, BRRI dhan39, BRRI dhan40, BRRI dhan41, BRRl dhan42, BRRI dhan43, BRRI dhan45 and BRRI dhan46) were sterilized by 70% ethanol for three minutes with gentle shaking followed by 0.10% mercuric chloride for 20 minutes. Sterilized seeds were then washed for 5-6 times with sterilized distilled water and dried on sterilized filter paper. Media were prepared for callus induction based on the formulation of MS basal salts supplemented with 0.8% agar, 20 g/L sucrose and 2 mg/L 2, 4-D. Five different concentrations (1.0,2.0,3.0,4.0 and 5.0 g/L) of two salts (NaCI and Na₂SO₄) were used in the media to evaluate their effects on germination and callus induction. Eight sterilized seeds were plated on each petri dish and 768 seeds were used for each variety for each treatment. The seeds were incubated for 28 days at 27^OC (± 1^oC) under dark. For regeneration. MS medium supplemented with 0.8% agar, 40 g/L sucrose, I mg/L NAA and I mg/L Kinetin was used for plant regeneration. Four weeks old calli were transferred into test tubes (25 x 200 cm ) containing 8-10 ml regeneration medium. Data on percent seed germination, percent callus induction and percent regeneration were calculated.
