Colliform Based Quality of Vola Fish (johnius Argentatus) Available at Some Local Fish Markets of Khulna City

Abstract/ Executive Summary:

An investigation was carried out on the microbiological quality of Vola (Johnius argentatus) available at local fish markets of Khulna city. The microbiological quality was compared with recommended microbial limits of International Commission on Microbiological Specifications for Foods (ICMSF) and European Union (EU). The samples were collected from four local fish markets namely Newmarket, Moilaputa Evening Market, Gollamari Fish Market and Nirala Fish Market. Five microbiological attributes (Standard Plate Count, Total Coliform, Faecal Coliform, Salmonella sp and Vibrio cholerae) were assessed according to the Bacteriological Analytical Manual of the United States Food and Drug Administration. The study was done in February 2008 in the Fish Quality Control Labratory of Fisheries and Marine Resource Technology Discipline of Khulna University. The results varied market-wise but all the microbiological qualities were found within the acceptable limits recommended by ICMSF and EU. The SPC varied between 0.44x105 cfu/g to 5.35x105 cfu/g. Total coliform counts varied between 4 to 43 MPN/g. Faecal Coliforms were found within the range <3 to 11 MPN/g. No Vibrio cholerae or Salmonella sp were found in any of the samples. The overall microbiological quality of Vola (Johnius argentatus) was good.

Objectives:

1. The present study was done with a view to observing whether the microbiological qualities of Vola available in the local fish markets of Khulna city are within the acceptable limits as recommended by International Commission of Microbiological Specifications for Foods and European Union.

Methodology:

Sample Collection and Preparation - The Vola fish were collected from four local fish markets of Khulna city namely Newmarket, Moilaputa Evening Market, Gollamari Fish Market and Nirala Fish Market. Microbiological Analysis - Microbiological analyses were done according to the Bacteriological Analytical Manual of the United States Food and Drug Administration. The laboratory tests were performed in the Fish Quality Control Laboratory of FMRT Discipline of Khulna University. All the tests were replicated for thrice.

Achievement/Findings:

Current investigations were carried out to study the Coliform based microbiological quality of Johnius argentaus available at New market, Moilapota evening market, Gollamari fish market and Nirala fish market of Khulna city encompassing both frozen and fresh samples. The microbiological quality was then compared with recommended microbial limits for fresh and frozen fish by International Commission on Microbiological Specification for Foods (ICMSF) and European Union (EU). Based on SPC, TC, FC assessment with no Vibrio cholera and Salmonella sp. detection, it was evident that the microbiological limits by the edible vola fish available at these local markets lied in these recommendations of ICMSF and EU. Though the microbiological quality of all the samples were within the recommended limit but quality can be maintained for long time if the proper icing is done to reduce bacterial growth and more hygienic condition is maintained. However, there were some limitations in the study. Only once the samples were collected from the markets. For more accurate scenario about the total microbiological quality, detailed survey with large number of samples and at different selling stages are recommended.
