In Vitro Cormlet Production of Gladiolus

Abstract/ Executive Summary:

Flower stalk and corrnel-tip explants were tested for in vitro propagation of gladiolus in cIutivar 'Carnival'. The nodal segements of flower stalk produced higher number of shoots when cultured on MS medium supplemented with 10 µM BAP, Shoot length was also highest at this concentration, Regenerated shoots produced roots and induced large cormlets at the basal end when 20 µM IBA and 60 g/l sucrose were added to half MS medium. In vitro cormlets germinated in the field and produced flowers.

Objectives:

The present investigation was undertaken to develop a protocol for large scale cormlet production of gladiolus.

Methodology:

Gladiolus cv. Carnival, grown at the Floriculture Farm of Bangladesh Agricultural Research Institute, Joydebpur, Gazipur was used as the source of explants. Collected explants were rinsed with tap water for 30 minutes. In case of cormel tip, the brownish covers were removed by scalpel and excised the inner cormel tip. Flower stalks were cut into pieces with nodal segments. Flower stalk and cormel tip segments were kept in different beakers and sterilized with 30% sodium hypochlorite solution for 20 and 30 minutes, respectively. After sterilization, explants were rinsed three times in sterile distilled water. Then the prepared explant were aseptically cultured on MS medium supplemented with six concentrations of BAP (0, 1.0, 2.5, 5.O, 10.0 and 15.0 µ1M). The cultures were incubated in a growth room at 24 ± 1⁰C with 16 h photoperiod. The pH of the media was adjusted to5.8 before autoclaving. The materials were repeatedly subcultured in every 4-5 weeks interval on the same fresh medium to produce multiple shoots. Total number of shoots per explant and average length of shoots were recorded up to 5^th subculture. After 5^th subculture, the proliferated multiple shoots were separated with 6 to 8 shoots and placed in the half MS liquid media containing 6% and various concentration of IBA viz. 0, 10, 20 and 30 µM. Cormelts produced in cluster and harvested/separated and transferred to the soil for germination and flowering.
