Histopathological Observations of Radiation Damage in Early Embryonic Developement of the Hide Beetle, Dermestes Maculatus Degeer (coleoptera:dermestidae)

Abstract/ Executive Summary:

Histopathological observations of gamma irradiation following early embryonic development of D. maculatus were studied to evaluate the feasibility of sterilizing the beetle as eggs for the application of Sterile Insect Technique (SIT). The developmental stages of D. niaculutus were recorded in different time sequences. In the control zygote nucleus, cleavage of the egg, blastoderm formation and the germ band initiated in the early embryonic development. The zygote nucleus, cleavage nuclei and the blastoderm stages of the beetle were found to be sensitive to radiation damage while the germ band stage was visibly radio- resistance at the lower dose levels of 10-50 Gy. In general, it was observed that irradiation doses of 10-50 Gy had a profound effect on the embryonic development of D. niaculatus .

Objectives:

To evaluate the feasibility of sterilizing the beetle as eggs for the application of Sterile Insect Technique

Methodology:

Dermestes maculatus was reared on dried kachki fish (Corica soborna, Hamilton) in the laboratory of Pest Ecology and Biology Division, Institute of Food and Radiation Biology, Atomic Energy Research Establishment, Savar, Dhaka, at 30±1°C and 70+5% R. H. in an incubator. The eggs of D. maculatus were collected from the rearing petridishes following different time sequences. The incubation period of the eggs was 54 hr. The eggs of 4, 12 and 16 hr were treated with doses of 10, 30 and 50 Gy at a dose rate of 1650 Gy/hr. The treated and untreated eggs were placed in seperate petridishes. The eggs were then fixed in alcoholic Bouin's fluid for 18-20 hr. The fixed eggs were washed in fresh 70% alcohol, They were then kept in 80% alcohol for 1hr and phenol solution for about 2hr and transferred to 90% alcohol for 1hr. Tertiary butyl alcohol was used to avoid hardening of the egg. The processed eggs were kept in this medium for about 2hr. It was then drained-off and eggs were kept in benzene for about 2hr. Embedding was done in paraffin wax in the usual way. The sections were cut at 6-8µm and were stained with Delafield's haematoxylin and counterstain with alcoholic eosin.

It may be concluded here that in order to find out proper dose for sterilization, its definite response to particular embryonic developmental stages of different insect species to radiation should be thoroughly studied for efficient use of SIT programme.
