Microbial Quality of Shrimp in New Moon and Full Moon

Abstract/ Executive Summary:

The present study was conducted to assess the microbial quality of shrimp in processing industry. Variations in quality of both raw and processed shrimps in lunar month were investigated for six months in terms of microbial load viz., SPC, TC, FC, Salmonella sp. and V. cholera. The microbiological tests were done during new moon and full moon and the results compared reveal the fact of SPC, TC and FC count being relatively high in full moon than in new moon. The SPC of shrimp at different sampling points were found ranging from 0.854 x 105 to 5.74 x 105 cfu/g during full moon and 0.614x105 to 4.8x105 cfu/g during new moon. The TC of shrimp varied between 7.80 to 40.20 MPN/g and 5.60 to 31.20 MPN/g in full Moon and new Moon respectively. The FC of shrimp were between 0.0 to 6.2 MPN/g in full moon and 0.0 to 5.4 MPN/g in new moon. Salmonella sp. and Vibrio cholerae were detected in no sampling points for both periods. The reasons of higher microbial load in full moon were ascribed mainly due to the yield of raw shrimp being higher than the carrying capacity of the plant and delay processing during this period. However, the loads found in shrimps of the processing plant were in satisfactory limit, which guarantee the acceptability of the plant products.

Objectives:

1. To assess the microbial quality of shrimp in processing industry during new moon and full moon to find out whether there is any variation in quality in lunar month with cause and resolution.

Methodology:

Study Area and period - There are 44 running shrimp processing plant in Khulna region. Among them 42 are EU approved. One of the EU approved processing plant was selected as study area. The study period was conducted over six months (June to November 2008). Microbiological Test of Shrimp - Microbial analyses followed the procedures of ICMSF. Plate count Agar (PCA)- 2.35g of plate count agar was first added in 100 ml of distilled water and boiled to dissolve the medium completely. The sulphite medium was then sterilized by autoclaving at 15l bs/inch2 pressure (121^oC) for 15 minutes. Lauryl Sulphate Tryptose Broth (LSTB) - 3.56g of LSTB was dissolved in 100 ml of distilled water and then was distributed into test tubes with insertingDurham tubes and sterilized by autoclaving at 15 lbs/inch2 pressure (121^oC) for 15 minutes.Durham tubes and sterilized by autoclaving at 15 lbs/inch2 pressure (121oC) for 15 minutes. Brilliant Green Lactose Bile broth (BGLB) 4g of BGLB was suspended in 100 ml of distilled water. It was mixed well and distributed in fermentation tubes and sterilized by autoclaving at 15 lbs/inch2 pressure (115oC) for 15 minutes. Salmonella sp. Culture media - Selenine Cystine Broth, Part-A - Tryptone –5g/liter, Lactose – 4 g/ liter, Sodium – 10 g /liter. Part- B - Sodium acid Selenine -4 gm/liter. 0.2g of part A and 1.33g of part B of selenite broth was suspended in 100 ml distilled water. It was warmed to mix well and dispensed and sterilized in boiling water bath or in free flowing steam for 10 minutes. It was not kept in autoclave because excessive heating is detrimental. Vibrio cholerae - Culture media - Alkaly Peptone Water (APW) - 4.5g of Alkaly Peptone Water (APW) was suspended in 225 ml distilled water and slightly heated to dissolve completely and sterilized by autoclaving at 15 lbs/ inch2 pressure at 20 minutes. Thio-sulphate Citrate Bile Salts Sucrose Agar (TCBSA) - 4.45g of Thio-sulphate Citrate Bile Salts Sucrose Agar (TCBSA) was first boiled in 50 ml of distilled water to dissolve and boil for 1 minute. Then the media was dried to 60oC for 15 minutes and was poured into petridishes and allowed to solidify.
