Culture of Rotifer, Brachionus Plicatilis Under Different Conditions Fed on Baker's Yeast

Abstract/ Executive Summary:

Rotifer, Brachionus plicatilis isolated from a semi-intensive shrimp farm and successively raised under laboratory conditions were used as seed to culture it under variable salinities and different source of salt water. The effect of initial stocking density on population growth, fecundity under variable salinity conditions and the reducing size of rotifers high density culture were also investigated. B. plicatilis was found to grow in a wide range of salinity from 5-35 ppt and highest growth was recorded under 30ppt (198 animals/ml). Initial higher rate of inoculation of seeds (20 animals/ml) showed relatively higher population growth of router (362 individuals/ml) at the end of 25 days of rearing. Routers stocked at 15-35ppt salinities supported adult population having relatively high fecundity (25-31%) and high density rotifer population (355 animals/ml) reduced the average size of the animal from initial average of 16µ to 90µ. Rotifers raised with Identical salinity of 25ppt under normal sea water, commercial NaCI, unpolished NaCI and brine solution yielded comparable population of 142 to 155 animals/ml within 21 days.

Objectives:

To look into population growth of rotifer under different salinities, and size variation of B. plicatilis under high density culture.

Methodology:

The experiments were carried out at FRI Marine Fisheries and Technology Station. Cox's Bazar during 1992-95. Rotifers, B. plicatilis were isolated from a pond of a shrimp farm. Animals isolated by screening methods were raised in 20cppt salinity in three polycarbonate tanks of 100 litre capacity each. Baker's Yeast was used as a single teed and the culture continued for 69 days. Occasionally some rotifers were discarded together with bottom water and sediments. Every day one third of culture water was discarded and replaced with 20ppt salt solution. Counting of rotifer were done by a stereo-microscope. Before counting a drop of formalin was added to sample on counting chamber to kill the rotifer which helped easy counting. Nine polycarbonate tanks of 100 litre capacity each were divided into 3 groups. Artificial salt water of 20ppt were filled up to 100 L mark. Rotifer produced in the laboratory as mentioned above were used as seed. For collection of seeds, aeration was stopped in culture tanks for few hours to deposit the suspended debris at bottom. Only swimming rotifers were harvested by suitable mesh. Rotifers were stocked at 10. 15 and 20 individuals/ml in 3 groups, respectively. Yeast were fed to rotifers twice a day depending on color or culture. Twenty four, flat bottom flasks (1L) were divided into 8 groups. Eight levels of salinity adjusted salt were taken into flasks with 3 replications. Identical number of containers were used for the experiment as explained in the preceding experiment, and all other culture conditions were also similar. Only difference was initial stocking rate and fecundity percentage, which was 40 animals/ml and 32% respectively for all trials. Laboratory cultured rotifers were filtered with 250, 160 and 120µ mesh successively. A stereo-microscope was used for counting, and micro-meter and electrical microscope was used to determine the size of rotifers. Only the size of egg bearing females and relatively large rotifers were determined to ensure that adults were measured. So far all preceding experiments were done with artificial salt water.

This investigation showed that commercial salt, brine solution or unpolished salt can be used for rotifer culture. For laboratory purpose, where there is no direct sea water supply, raising of B. plicatilis or maintaining the seed for future use can be done. The findings indicates that rotifers can be easily reared under the existing condition or the country for experimental as well as mass culture basis at low cost.
