Mechanical inoculation test proved that virus disease like symptoms found in china rose (Hibiscus rosa-sinensis) plant grown at the BSMRAU, Gazipur. It was sap transmitted. Healthy plants of china rose were inoculated with the purified isolates obtained from the local lesion isolation using Gomphrena globosa and Chenopodium amaranticolor. chlorotic spots, ring spots, leaf cupping and yellowing. The symptoms were appeared to be similar to those observed in inoculated plants as observed in the naturally infected plants. G. globosa and C. amaranticolor produced necrotic and chlorotic local lesions, respectively, when inoculated with 0.01M Potassium phosphate buffer, PH 7.0. Other than china rose, a member of Malvaceae family the virus was mechanically transmitted to the plant species belonging to family of Amaranthaceae, Chenopodiaceae, Cucurbitaceae and Leguminosae. Its dilution end point was determined as 10-8 while thermal inactivation point was 70C and longevity in vitro was 12 days. The virus did not react positively against the antisera of Cucumber mosaic virus, Papaya ringspot virus, Watermelon mosaic virus 2 and Zucchini yellow mosaic virus in Double Antibody Sandwich Enzyme-Linked Immuno-Sorbent Assay and Potato virus Y in Indirect Enzyme-Linked Immunosorbent Assay, although it infected the plants of Cucurbitaceae upon mechanical inoculation. Based on Symptomatological study and the results of the tests described above it was concluded that the virus found in naturally infecting china rose might be Hibiscus chlorotic ringspot virus (HCRV).

To identify the causal agent of virus disease-like symptoms in china rose and to find out the crop hosts of the causal agent infecting china rose.

The symptoms appeared in the naturally infected China rose plants were closely and carefully observed. The symptoms developed in the foliar parts of the plants including leaves and flowers were noted. The photographs of the infected plants were also taken for further illustrations. The china rose plants grown at BSMRAU campus having virus disease- like symptoms were observed. One of the infected plants was selected for the collection of samples. The leaves showing clear virus disease like symptoms were collected in polythene bags and carried to the laboratory for processing and further use. The collected samples were preserved as it was in a –30°C freezer and 4°C in the plastic petridishes containing silica gel. A piece of blotter paper was put in each plastic petridishes to spread the cut sample on to it. The plastic petridishes were then wrapped with scotch tape to make it air tight. The mechanical inoculation test was carried out following the method described by Hill. Three different plant species were used in the test. The plant species were Hibiscus rosa-sinensis, Chenopodium amaranticolor and Gomphrena globosa. The plants were grown in an insect proof nethouse using five inches earthen pot. The soils used in the pot were amended with manure and fertilizers as per recommendation. Proper care including irrigation was done to confirm the normal growth of the plants. In case of C. amaranticolor and G. globosa seedlings were transplanted in the pots containing sterilized soil. Their seedlings were grown in plastic tray inside the nethouse. Apparently healthy cuttings were planted in pots containing sterilized soil.

China rose (Hibiscus rosa-sinensis) were found to be infected with a virus producing virus disease- like symptoms such as chlorotic ring spots, yellowing and cupping of leaves. It was mechanically transmissible. The virus induced characteristics local lesions in Gomphrena globosa and Chenopodium amaranticolor. Other than Malvaceae family the plant species belonging to Amaranthaceae, Chenopodiaceae, Cucurbitaceae and Leguminosae were found to produce symptoms upon mechanical inoculation. However, the plant species belonging to Compositae, Cruciferae, Gramineae and Solanaceae did not have any response in inoculation test. The dilution end point, thermal inactivation point and longevity in vitro were determined as 10-8, 70°C and 12 days at room temperature (30 ± 2 °C), respectively, using crud sap extract of systemically infected Hibiscus rosa-sinensis. In Double Antibody Sandwich Enzyme-Linked Immuno-Sorbent Assay and Indirect Enzyme-Linked Immunosorbent Assay the virus did not react positively against the antibodies of Cucumber mosaic virus (CMV), Papaya ringspot virus (PRSV), Watermelon mosaic virus 2 (WMV-2), Zucchini yellow mosaic virus (ZYMV) and Potato virus Y (PV Y). The virus under the study had similar characteristics as observed in HCRV infected Hibiscus rosa-sinensis plants.
