Soil salinity is one of the major environmental stresses affecting the rice growth and reduces the grain yield significantly. In vitro production of double haploid plants through anther culture provides an efficient system for rapid production of homozygous lines. Therefore, attempt has been undertaken for developing suitable salt tolerant rice variety through anther culture. Boots were collected at appropriate stage from F1 plants and pre-incubated at 8ºC for 8 days. The panicles were then cut into small pieces and surface sterilized by 70% ethanol. For callus induction, anthers were incubated on two different media (KE and M10) and kept in an incubator under dark condition at 25ºC.  Induced calli were then transferred into regeneration medium (MS medium containing 1.0 mg/l NAA+ 1.0 mg/l Kinetin) for regeneration. Data were taken on number of anther plated, number of calli produced, number of albino and green plants regenerated. A total of 62657 hybrid anthers of 27 F1 populations were plated in KE and M10 media for callus induction. A total of 330 calli were obtained from KE medium and 10 calli were obtained from M10 medium. The highest numbers (73) of calli were obtained from hybrid anthers of BRRI dhan29/MR219 in KE medium. However, nine green plants were regenerated from hybrid anther of BRRI dhan29/FL478 and three plants from MR219/IRBB60, respectively. After hardening, green plants were transferred into the earthen pots and seeds were harvested at maturity. Harvested seeds will be used for further evaluation and screened for tolerance to salinity.

To develop salt tolerant rice varieties.

Twenty seven F1s populations were grown in the net house under optimum management. Boots with appropriate stage (uninucliate/mid uninucleate at reduction division stages) were collected from F1 plants. First the boots were pre-incubated in a refrigerator at 8ºC for 8 days. The panicles were then taken from the boots and cut into small pieces. They were then surface sterilized by 70% ethanol for 2-3 minutes and then placed onto the sterile filter paper in Petri dishes to remove excess alcohol. For callus induction, anthers were incubated into two different media (KA an M10) media and kept in an incubator under dark condition at 25ºC until cullus initiation. Calli were then transferred into regeneration medium (MS medium containing 1.0 mg/l NAA+ 1.0 mg/l Kn), and culture were then incubated in a culture room under cool white Fluorescent lamps (approximately 1000 lux, under 25ºC) for regeneration., Data were taken on number of anther plated, number of callus produced, number of green and albino plant regenerated.
