Studies on Ultimate Fibre Cells of Interspecific Hybrids Between Hibiscus Radiatus and Hibiscus Cannabinus L.

Abstract/ Executive Summary:

Ultimate fibre cells of F₁ and F₂progenies of the cross, Hibiscus radiatus X H cannabinus along with their parents were studied. In the two parents and in the F₁, F₂ generation fibres were located in the middle portion of the stem the next longest was found in the top portion and shortest at the basal portion.

Objectives:

To determine the fibre quality of H. cannabinus. H. radiatus, their F₁s and selected resistant F₂ lines before advancing generations with thus interspecific hybrid derivative lines.

Methodology:

Hibiscus cannabinus L. (cultivated variety), Hibiscus radiatus L. (wild) and their F₁, F₂ (from the crosses of H. radiatus and H. cannabinus) were used in the present investigation. The experiment was conducted in the field and laboratory of Bangladesh Jute Research Institute. The F₁, F₂ and parental seeds were sown in late March in randomized complete block design with two replications. Standard cultural practices were maintained. Materials of the same age, height and stem diameters were taken. For studying the ultimate fibre cells, portion of barks (5 cm in length) were collected from base, middle and top portion of the stem at average flowering time and fixed in FAA. Washed in distilled water with 3-4 changes. The bark was scrapped with scalpel to remove periderm. The material was then boiled in a retting solution comprising of 1gm caustic soda, 2gm ammonium oxalate, 4gm sodium sulphate, 10 drops of concentrated nitric acid and 100ml of distilled water for 15 minutes. After boiling, it was washed with distilled water, gummy substances was removed again with scalpel. The materials were again boiled in fresh macerating solution for 15 minutes. After washing and cleaning it was again boiled first in distilled water for 10 minutes and then in 5% chromic acid for 3-5 minutes. After washed thoroughly, the materials were then ready for ultimate fibre cell studies. Single strand of fibre was separated by fine needles, stained with 2% safranin and temporarily mounted with 10% glycerine for microscopic study. Each of the ultimate fibre was taken and the measurements were taken from one end of the fibre to the other end. From the outer border of one side of ultimate fibre cell to the outer side of the other side of cell was taken for breadth measurement.

In the two parents and in the F₁, F₂ generation fibres were located in the middle portion of the stem the next longest was found in the top portion and shortest at the basal portion. In kenaf the middle portion showed the highest mean length of ultimate fibre cells. The top portion constitutes a small portion of the whole stern and if harvested before flowering, most of the fibres get lost during retting, while the middle portion of stem constitutes the major portion and does not have chance to get lost during retting. In this respect also, Kenaf is considered more suitable as raw materials for paper pulp.
