The research work was conducted using different concentrations of H2O2 in the substrate of oyster mushroom (Pleurotus ostreatus) for controlling associated mycoflora. Spawn packets were treated with five different concentrations of H2O2 viz. 5%, 4%, 3%, 2%, 1% and control spawn packet (without using H2O2 but using autoclave and laminar air flow). Occurrence of the lowest contamination 3.33% spawn packets were found in case of 3% to 5% H2O2 treated spawn packet. Minimum time (18.33 days) was required for completing mycelium running in the spawn packet and the fruiting bodies formations of oyster mushroom was required for control spawn packet nevertheless, 3%, 2% and 1% H2O2 treated spawn packets also required statistically similar minimum time. The highest yield of fruiting bodies (680.50g) per spawn packet was observed in 3% H2O2 treated spawn packet, which is significantly higher than that of other H2O2 treated packets.

1. The present studies were undertaken to find out the appropriate concentration of H2O2 for controlling the associated mycoflora of oyster mushroom substrate.

The experiments were conducted at the laboratory of the department of Plant Pathology, Patuakhali Science and Technology University, Dumki, Patuakhali, Bangladesh. Collection of mother spawn of Pleurotus ostreatus - “PO2” isolate of Oyster mushroom (Pleurotus ostreatus) was selected to conduct the present study. Mushroom seed commonly named “mother spawn” of PO2 was collected from the National Mushroom Development & Extension Centre, Department of Agricultural Extension, Sobhanbag, Savar Dhaka, Bangladesh. Preparation of spawn packets - Substrate was prepared by mixing sawdust and wheat bran at the ratio of 2:1. Calcium carbonate was used at the rate of 0.2%. The moisture level of the substrate mixture was adjusted approxymately 65% by adding water. Here water with different concentrations of H2O2 viz. 5%, 4%, 3%, 2% and 1% were added with substrates according to treatments. 10 spawn packets for each treatment with 3 replications were taken to evaluate occurrence of contamination. The spawn packets were prepared separately with each type of substrate. Polypropylene bags of 22.5 cm X 30 cm size were filled with 500 g of prepared mixture and packed tightly. All packets were labeled with required information. The neck of the bag was prepared by using plastic heat resistant neck. A hole of about 2/3 of the volume of the bag was made with a sharp end stick at the center for space to put inoculums. The neck was plugged with cotton and covered with brown paper by placing a rubber band to hold it in place. Inoculation with Pleurotus ostreatus was done by the mother spawn at the rate of two teaspoonfuls per packet. Then the inoculated packets were placed on rack of incubation chamber at 25 ± 20C temperature. Evaluation of H2O2 against Growing spawn packet - The brown paper, rubber band, cotton plug and plastic neck of the spawn packets were removed and the mouths of polypropylene bags were wrapped and tied tightly with rubber bands. Two ends, opposite to each other of the upper position of the plastic bag were opened by removing the plastic sheet with a scalpel making ‘D’ shape opening. The opened surface of the substrate was scraped slightly with a teaspoon for removing the thin whitish mycelial layer. The spawn packets were soaked in water for 15 minutes and inverted to remove excess water for another 15 minutes. The packets of each type were placed separately side by side on the rack of culture house. The relative humidity and temperature of culture house were maintained in higher and lower level, respectively by watering thrice daily. Diffused daylight and proper ventilation in culture house were also maintained for fruiting body development. The mushroom fruiting body was harvested by gentle twisting before the mushroom show slightly curled edges. After first harvest the scrapping and soaking were done as first opening of the packet but after the second and the successive harvests only scrapping was done. Watering and other activities were continued as before. Data collection - Data on the parameters of number of fruiting body, Weight of fruiting body, diameter of pelius and thickness of pelius were collected.

Effect of H2O2 on different yield attributing characters including number and weight of fruiting bodies, diameter of pileus and total yield of oyster mushroom were studied and the results are presente. Different concentrations of H2O2 treated spawn packets had a significant influence in yield contributing characters and total yield of mushroom. The number of fruiting body grown on different treated spawn packets differed significantly. The significantly highest mean number of fruiting bodies per packet was recorded 81.66 in 3% H2O2 treated spawn packet followed by 2% H2O2 treated spawn packet (80.33 fruiting bodies). In case of other two yield contributing parameters i.e. number of fruiting body and diameter of pileus showed similar result that 3% H2O2 treated spawn packet (81.66) performed better. The highest yield of fruiting bodies per spawn packet was observed 680.50g in 3% H2O2 treated spawn packets which was significantly higher than that of other H2O2 treated packets.
