Development of High Yielding, Disease Resistant and Salt Tolerant Rice Lines Through Anther Culture

Abstract/ Executive Summary:

Soil salinity is one of the major environmental stresses affecting the plant growth adversely and reduces the grain yield significantly. About 1.0 million hectare of cultivable land in Bangladesh is affected by various degrees of soil salinity. Therefore, attempts have been undertaken for developing suitable salt tolerant variety through anther culture.A total of 86922 hybrid anthers of 29 (cross combinations) F₁populations were plated in KE and M₁₀media for callus induction to develop salt tolerant lines.A total of 131 calli were obtained from KE medium and 16 calli were obtained from M₁₀ medium.The highest numbers (24) of calli were obtained from hybrid anthers of BRRI dhan29/MR219 in KE medium.Calli obtained from induction medium were transferred in to the regeneration media and only ten green planlets were regenerated from hybrid anther of BRRI dhan29/FL47 and MR219/IRBB60. Green plants were put into nutrient solution for hardening and finally they were transferred into the earthen pots and kept there until maturity.

Objectives:

To develop high yielding, early maturity and salt tolerant rice variety.

Methodology:

For this study, 29 F_1s populations were grown in the net house under optimum management. Boots with appropriate stage (uninucliate/miduninucleate at reduction division stages) were collected from F₁ plants. First the boots were pre-incubated in a refrigerator at 8ºC for 8 days. The panicles were then taken from the boots and cut into small pieces. They were then surface sterilized by immersing 70%ethanol for 2-3 minutes and then placed onto the sterile filter paper in Petri dishes for emove excess alcohol. For callus induction, anthers were incubated into two different media (KA an M10) and kept in incubator under dark condition at 25ºC until cullus initiation. Calli were then transferred into regeneration medium (MS medium containing 1.0 mg/l NAA+ 1.0 mg/l Kn), and culture were then incubated in a culture room under cool white Fluorescent lamps (approximately 1000 lux, under 25ºC) for regeneration. Eight weeks after transfer of calli into the regeneration medium, data were taken on number of anther plated, number of callus produced, number of green and albino plant regenerated.
