Introgression of Sub1 Gene Into BRRI Dhan44 Through Marker Assisted Backcross Breeding

Abstract/ Executive Summary:

BRRI dhan44 is a popular variety in the tidal areas of Bangladesh due to its tall stature and bold grain shape. But sometimes it is victimized by submergence for about two weeks when high tides occur and water mass intrudes cultivated lands and eventually the plants die. For this reason some farmers in that area prefer local varieties overlooking their low yield potentiality as they show a bit higher tolerance. It would be a great deal to introgress sub1 gene into BRRI dhan44 to popularize it more in that area as well as the other areas of the country where the variety is cultivated and subjected to inland submergence. And marker assisted backcrossing is preferred for it potential to recognize the precisely introgressed plant samples.Seventy five polymorphic markers were used in BC₃F₁ and BC₄F₁ generation for background selection of BRRI dhan44.Five homozygous plants of BC₄F₁ were selected for backcrossing.Foreground selection was done in BC₅F₁ generation and heterozygous plants were selected with SUB1 C173 primer .In BC₅F₂ plants,foreground selection was done again with SUB1 C173 primer to confirm homozygous plants SUB1 gene in BRRI dhan44.Forty four plants were identified for further evaluation.

Objectives:

To make the variety BRRI dhan44 submergence tolerant for about two weeks.

Methodology:

BRRI dhan52 was used as the donor parent for sub1 gene and the BRRI dhan44 was used as the recurrent parent. Both parents were crossed to generate F₁ population and then backcrossed. Primer survey was done to select the polymorphic markers which are distributed throughout the genome. This data were used for background selection afterwards. Foreground selection was done in the BC₁F₁ and BC₂F₁ population to select the plants possessing the target gene and back crossed. Best plants within the backcross populations were selected phenotypically. In this way BC₃F₁ population was generated and background selection was done. On the basis fbackgroundselection,homozygous plants of BC₄F₁ were selected for backcrossing. Foreground selection was done in BC₄F₁ and BC₅F₁ generations and heterozygous plants were selected with SUB1 C173 primer. In BC₅F₂ generation, foreground selection was done again with SUB1 C173 primer to confirm SUB1 gene in BRRI dhan44.
