Crescentia cujete and Moringa oleifera leaves were tested for antibacterial activity against nine pathogenic bacteria using Agar-Cup method. The chloroform extract of Moringa oleifera exhibited the largest zone of inhibition (29.3 mm) at a concentration of 8 mg/well against Shigella dysenteriae. Ethanolic extract of Crescentia cujete showed highest zone of inhibition (23.35 mm in diameter) against Shigella dysenteriae at a concentration of 8 mg/well. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined by broth macrodilution method. The lowest MIC (2500 μg/ml) and MBC (3000 μg/ml) of crude ethanolic extracts of Crescentia cujete was recorded against Bacillus subtilis. In case of Moringa olifera the lowest MIC (1500μg/ml) and MBC (2000μg/ml) was with the crude chloroform extracts against Bacillus megaterium and Bacillus cereus respectively. The regression (R2) value of extract concentration with zone diameter varies from 0.850 to 0.990 for Moringa oleifera and 0.806 to 0.997 for Crescentia cujete. Our present study suggests that active antimicrobial agents present in the extracts of Crescentia cujete and Moringa oleifera may have potential for the treatment of bacterial infection.

1. To test antibacterial activity of Crescentia cujete and Moringa oleifera against nine pathogenic bacteria using Agar-Cup method.

Collection and extraction of plant material - The leaves of Crescentia cujete were collected in fresh condition from Gazipur and Bangladesh Agricultural University, Mymensingh; and Moringa oleifera leaves were collected from BCSIR campus. The cleaned samples were cut into small pieces (1-2 cm), dried in air to make it suitable for grinding. The samples were ground to fine powder mechanically and 20 g dried powder was kept steeped 72 hours in Chloroform, carbon tetrachloride and petroleum ether separately. On the other hand, 250g of the dried powder were kept stepped 72h in ethanol. The extract thus obtained was filtered and subjected to rotary vacuum evaporator at 50º C and concentrated to gummy materials under reduced pressure. The gummy materials were then collected in a small vial and then dried. Thus crude extracts were obtained. Determination of antibacterial activity - The extracts of both plants were tested for antibacterial activity against nine pathogenic bacteria viz., Shigella dysenteriae CRL, (ICDDR,B), Salmonella typhi CRL, (ICDDR.B), Salmonella paratyphi CRL, (ICDDR,B), Escherichia coli ATCC 8739, Staphylococcus aureus ATCC 25922, Bacillus subtilis Prof. Skersman, Australia, Bacillus megaterium Prof. Skersman, Australia, Bacillus cereus Prof. Skersman, Australia and Sarcina lutea ATCC 9341. Bacterial inoculums were prepared by Clinical and Laboratory Standards Institute (CLSI) guideline. Bacterial cultures were emulsified in normal saline and turbidity was matched with 0.5 McFarland turbidity standards. The agar cup method (Barry, 1980) was followed to investigate the antibacterial activity of the extracts. 0.1 ml of TSB broth culture of the test organisms were firmly seeded over the Mueller Hinton Agar plates. Wells of 6 mm diameter was punched over the agar plates using a sterile cork borer. The bottoms of the wells were sealed by pouring 50 - 100 μl of molten MHA into the scooped out wells. Using a micropipette, extracts of different solvents were added to different wells in the plate. These plates were then kept at low temperature (4ºC) for 2-4 hours and incubated at 37 ºC for 24 hours. After the incubation period formation of zones around the wells, confirms the antibacterial activity of the respective extracts. All the results were compared with the standard antibiotic disc of Doxycyclin Hydrochloride. Determination of MIC and MBC - The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values of crude extracts against the test pathogens were determined by micro and macrodillution broth technique using Mueller-Hinton medium.

The MIC values of M. oleifera were found varied from 1.5 mg/L to 2.5 mg/L against test pathogens. The chloroform extract showed the lowest MIC value (1.5mg/L) against Bacillus subtilis and Shigella dysenterieae. The MBC values were found between 2 mg/L to 3 mg/L against the test pathogens. The chloroform extract exhibited the lowest MBC value (2 mg/L) against Bacillus subtilis and Shigella dysenterieae.
