Mapping of Ufra Resistant Gene

Abstract/ Executive Summary:

Disease is one of the most important factors which hinders the yield of rice plant. Among the major diseases ufra affects the rice field in all the three rice growing season and yield loss due to this disease may be 100 percent under favorable environment condition. Chemical control which is effective against this disease to some extent costly and also not environmentally sound. Resistant variety is the most sustainable technology to combat diseases. Gene mapping can identify the exact position and linkage groups of the genes for resistant to ufra diseases. Information about the positions of the resistant genes and the molecular markers linked to them will be useful for marker-assisted selection in ufra resistant rice breeding. One hundred individuals (50 highly susceptible and 50 highly resistant) of F₂ population selected from cross, IR71362-3B-8-3-1-2B / R16-06-3. IR71362-3B-8-3-1-2B is a HYV susceptible while R16-06-3 is an ufra resistant line. Leaf samples from selected individuals along with parent were collected at 30 DAS for DNA extraction and those lines were transplanted in the field. DNA extraction were done following modified CTAB method. Phenotypic study were done in same F₂generation following inoculation method of 100 nematodes/ plant. For genotypic study, collected DNA was amplified using SSR primers and run electrophoresis. The banding pattern was visualized by staining in ethidium bromide and documented. Linkage analysis was performed by using the Map Manager/QTX computer program (Manly et al. 2001). One hundred and three primers were surveyed and thirteen were found polymorphic.

IR71362-3B-8-3-1-2B is a HYV susceptible while R16-06-3 is an ufra resistant line.
