Establishment of in Vitro Plant Regeneration System from BRRI Released Rice Varieties.

Abstract/ Executive Summary:

Low rates of callus induction and subsequent plant regeneration with Indica rice are major drawbacks in in vitro culture. The influence of media composition on callus and regeneration are being reported by various authors. Therefore, the effect of different media on callus induction and subsequent plant regeneration from mature seed embryos of BRRI released rice varieties was tried. Highest percentage (100%) of germination was found in several varieties viz. BR16, BR23, BRRI dhan36, BRRI Hybrid1 and Taipei309 (control) in different media. Lowest germination was found in BR2 (5.0%) followed by BRRI dhan38 (6.67%) in N6 and MS media, respectively . The study revealed that media composition had tremendous effect on callus induction frequency. The BRRI varieties showed diverse result with changes of the medium. The highest callus induction frequency was found in BRRI dhan48 (48.33%) with MS media which is similar to control Japonica rice Taipei 309 in MS and N6 media. No callus was recovered from several test varieties in different callus induction media, like BRRI dhan42 produced no callus on N6 media but 3.33% callus was developed on MS and Nitsch & Nitsch media.

Objectives:

To identify the best culture media for callus induction and subsequent plant regeneration of BRRI released rice varieties.

Methodology:

BRRI released 49 inbred (BR1 to BRRI dhan50) and two hybrids (BRRI hybrid dhan1 and BRRI hybrid dhan2) were used in this study. Japonica rice Taipei 309 was included as a control. First mature healthy dehusked seeds were sterilized by 2-3 drops of Tween20 for 10 minutes. Seeds were then sterilized with 70% ethanol for 1.5 minutes followed by 0.1% mercuric chloride for 15 minutes. After then sterilized seeds were washed 5-6 times with sterile distilled water. Then the sterilized seeds were plated in Petri dish containing three different callus induction media (MS, N6 and Nitsch & Nitsch) supplemented with 2.0mg/L 2,4 dichlorophenoxy acetic acid (2,4-D) and gelled with 0.3% Phytagel. Petri dishes were sealed with parafilm and incubated for three weeks at 27±1⁰C under dark condition before callus induction responses were assessed. After three weeks of incubation data were taken on germination and callus induction frequency. For regeneration, four-week-old embryogenic calli were placed into the test tubes containing three different regeneration media (MS, N6 and Nitsch & Nitsch) supplemented with 1.0mg/l NAA + 1.0mg/l Kinetin. Cultures were kept at 25±1⁰C under continuous light. Data were taken for shoot regeneration four weeks after transferring the calli into the regeneration medium.
