Effect of Hot Water Treatments in Controlling Seed Borne Fusarium Moniliforme in Rice Seeds

Abstract/ Executive Summary:

An experiment was conducted at Plant Pathology Laboratory, BRRI, Gazipur to find out a suitable temperature and time of exposure to eradicate the seed borne F. moniliforme of rice seeds. There are three temperature and time of exposure were used. Seeds were soaked into hot water for different temperature and time of exposures. The findings reveal that germination of hot water treated seeds at 55^oC for 15 minutes at 40% higher than the non treated seed. All the temperature level and time of exposure significantly reduce the infection of F. moniliforme but not completely eradicate except ta 60^oC temperature. However at 60^oC temperature for 15 minutes F. moniliforme completely eradicate, but the germination rate reduced significantly. Vigour index of the treated seeds was higher at 55^oC for 15 minutes and than gradually decreases and lowest in 60^oC for 15 minutes.

Objectives:

To find out the possibility of hot water as an alternative of fungicides to control seed borne F. moniliforme

Methodology:

Sample Collection: Naturaly bakanae infected seeds of BR1 were collected from farmers field at Habiganj District during 2009. They were identified at seed health test with low (20%) Medium (20-40) and high (40%) infection of F. moniliforme at plant pathology laboratory, BRRI, Gazipur. Seed treatment and detection of seed borne fungi: Infected seeds were presoaked for five hours in sterile distilled water, than placed in cloth bags and immerse in water bath maintained at 50,55,60⁰C for 5,10 and 15 minutes.Bags containing the seeds after exposure to the desired temperature were placed in ice cold water for three minutes,blotted dry in filter paper and dried at 35^oC for one hour. Seeds were plated in replicates of 25 seeds per 9 cm diameter plastic petridish lined with three layers of water soaked blotter paper. Controlled plates were maintained with blotted dry seeds pre soaked for 5,10 and 15 minutes in sterilied distilled water. For each temperature level and period of exposure four replication were made. The petriplates were incubated at 30^oC for 9 days. The experiment was split-split-plot in CRD. After 9 days all plates were observed by stereomicroscope and prevalence of F. moniliforme and other fungi were recorded. Germination test: Germination of treated seeds was performed following the conditions described by ISTA (1999). Two hundred seeds were tested in four replications of 50 sees each, using blotted paper method. After rolling the blotted paper were placed in polythene bags and incubated at an upright position in a room temperature. The experiment was repeated twice. Vigour index test: The experiment was carried out in potted sterilized sand. Samples from untreated control and hot water treated seeds of different temperature and time of exposure were used. In each case 100 seeds were divided into 5 replications. The seeds were sown in earthen pot having 20 seeds/pot. The germination records were taken from days of germination starting and counted to completion of germination and vigour index was calculated following the formula of Agrawall (1996).

Achievement/Findings:

All the temperature level and time of exposure had significant effect on reducing the seed borne infection of F. moniliforme. Among the temperature levels seeds treated at 55^oC for 15 minutes was better. However at 60^oC temperature for 15 minutes F. moniliforme completely eradicate, but the germination rate reduced significantly. Vigour index of the treated seeds also higher at 55^oC for 15 minutes.
