Development of Salt Tolerant Rice Lines Through Anther Culture(2009-10)

Abstract/ Executive Summary:

Soil salinity is one of the major environmental stresses affecting the plant growth adversely and reduces the grain yield. Almost 1.0 million hectare of cultivable land in Bangladesh is affected by various degrees of soil salinity. Therefore, attempts have been undertaken for developing suitable salt tolerant variety through anther culture. Anthers of 12 F₁ populations were plated in KA, KE and M10 media. Highest numbers (18) of calli were obtained from hybrid anthers of BRRI dhan28 × BRRI dhan47 in KA medium followed by KE medium. Calli obtained from different induction medium were transferred into the regeneration media. Only Calli obtained from hybrid anther of BRRI dhan28 × BRRI dhan47 produced 14 green plants in KA medium.

Objectives:

To develop salt tolerant double haploid line through anther culture

Methodology:

For this study twelve F_1s populations were grown in the net house under optimum management. Boots with appropriate stage (uninucliate/mid uninucleate at reduction division stages) were collected from F₁ plants. The boots were preincubated in a refrigerator at 8ºC for 8-10 days. The panicles were opened from the boots and cut into small pieces. They were then surface sterilized by immersing in 70% ethanol for 2-3 minutes and then placed onto the sterile filter paper in Petri dishes to remove excess alcohol. For callus induction, anthers were incubated into two different media (KA and KE) and kept in an incubator under dark condition at 25ºC until callus initiation and subsequent growth up to appropriate size (appx. 2.0 mm diameter). Calli were then transferred into regeneration medium (MS medium containing 1.0 mg/l NAA+ 1.0 mg/l Kn). Cultures were incubated in a culture room under cool white Fluorescent lamps (approximately 1000 lux, under 25ºC) for regeneration. Eight weeks after transfer of calli into the regeneration medium, data were taken on number of anther plated, number of callus produced, number of green and albino plant regenerated.
