Development of Salt Tolerant Rice Line Through Anther Culture

Abstract/ Executive Summary:

Soil salinity is one of the major environmental stresses affecting the plant growth adversely and reduces the grain yield. Almost 1.0 million hectare of land in Bangladesh is affected by varying degrees of soil salinity. Therefore, attempts are undertaken to develop suitable salt tolerant variety through anther culture. Six F₁s were used for this study. Three F₁ population were grown under the optimum fertility and management. Boots with appropriate stage were collected and pre-incubated in a refrigerator at 8ºC for 8-10 days. The panicles were cut into small pieces. They were then surface sterilized, incubated in three different media (KA, KC and KE) and kept in dark condition at 25ºC and calli were then transferred into regeneration medium (MS medium containing 1.0 mg/l NAA+ 1.0 mg/l Kn)and incubated in white fluorescent lamps (approximately 1000 lux, at 25ºC) for regeneration. Highest number (33) of calli induction were obtained from hybrid anthers of BRRI dhan29 × BRRI dhan40 in KA medium followed by the same hybrid in KE medium (18) and BR4909R₁R₂ × BRRI dhan41 in KA medium (18). Only BRRI dhan28 X Pokkali gave 11 green plants.

Objectives:

To develop salt tolerant double haploid line.

Methodology:

For this study three F₁ population were grown in the net house under the optimum fertility and management. Boots with appropriate stage (uninucliate/mid uninucleate at reduction division stages) were collected from F₁ plants. The boots were pre-incubated in a refrigerator at 8ºC for 8-10 days. The panicles were opened from the boots and cut into small pieces. They were then surface sterilized by immersing in 70% ethanol for 2-3 minutes and placed onto sterile filter paper in Petri dishes to absorb access alcohol. For callus induction, anthers were incubated in three different media (KA, KC and KE) and kept in an incubator under dark condition at 25ºC until callus initiation and subsequent growth up to appropriate size (2.0mm). Calli were then transferred into regeneration medium (MS medium containing 1.0 mg/l NAA+ 1.0 mg/l Kn) and cultures were incubated in a cultured room under cool white fluorescent lamps (approximately 1000 lux, at 25ºC) for regeneration. Eight weeks after transfer of calli into the regeneration medium, data were taken on number of anther plated, number of callus produced, number of green and albino plant regenerated.

Anthers of six F₁ populations were plated in KA, KC and KE media. Highest number (33) of calli induction were obtained from hybrid anthers of BRRI dhan29 × BRRI dhan40 in KA medium followed by the same hybrid in KE medium (18) and BR4909R₁R₂ × BRRI dhan41 in KA medium (18). Calli of the F₁ populations obtained from different induction medium were transferred into the regeneration medium. Only BRRI dhan28 X Pokkali gave 11 green plants.
