The effect of three hormones (viz. PG, Ovaprim, and LHRH.A2) on the captive breeding of the exotic black carp, Mylopharyngodon piceus, was assessed at Arabpur Fish and Prawn Farm in Jessore district of Bangladesh during June-July 2000. Nine individual mature females ranging from 4 kg to 11 kg in body weight were used in three treatments (T1, T2, T3) each with three replications. Two injections of PG, in treatment-1 (T1) were administered to three individual females in two separate doses. The first and last dose were given at the rate of 3 and 7 mg/kg body weight, respectively. In treatment-2 (T2) a single dose of Ovaprim was given at the rate of 0.5 ml/kg body weight. In treatment-3 (T3) LHRH.A2 and PG were used in combinations. The first dose was given with LHRH.A2 only the rate of 1 μg/kg body weight. The second dose was given with a mixture of LHRH.A2 and PG at the rate of 4.5 μg and 3 mg/kg body weight, respectively. Two complete and one partial breeding in T1, three partial breeding in T2 and three complete breeding in T3 were observed. The highest latency period (11-14 hrs), fertilization rate (66%), hatching rate (92.67%) and rate of viable larvae (53.44%) were found with T3. Therefore, T2 (Ovaprim) was found to be the best hormone at the rate of 0.5 ml/kg body weight for the successful breeding of the black carp in hatchery condition.

The present study was conducted in Arabpur Fish and Prawn Farm, Jessore, during June-July 2000 to observe the effectiveness of three different hormone treatments in single and in combination of doses on the breeding of the black carp, M. piceus, in captive hatchery condition.  Nine gravid females were collected during spawning period (June-July) from the brood stock ponds and reared in control ponds at the above Farm, prior to conduct the experiment. The male and female fishes were acclimatized to hatchery conditions for 24 hours prior to the initiation of experiment in separate tanks. The male and female sexes were easily distinguished by observing pectoral fin characteristics. In male, the pectoral fin is long and strong with rough ventral side, while in female it is smooth. A mature male oozes milt freely by applying gentle pressure on the abdomen. A female fish with comparatively bulky abdomen and swollen, soft, reddish, elastic, genital opening were chosen for the experiment. Three breeding hormones were used in the present study in three separate treatments, viz. Pituitary Gland (PG) extracts, Ovaprim and LHRH.A2+PG extract. The solution was centrifuged for 15 seconds for precipitation with a hand centrifugate. The supernatant solution containing hormones was then ready for injection. The brood fishes were caught, wrapped with a soft and moist towel and injected intraperitoneally with the help of a 5 ml hypodermic syringe. After the male and female fishes being injected they were put into separate tanks. Spawning started 9-14 hours after the administration of first injection to the females. Water temperature was recorded with a Celsius thermometer. Dissolved oxygen (DO) and pH values were recorded by using a Portable Water Test Kit (Model FF-1A, HACH Company, Cat. no. 243002, Loveland Colorado, USA) and Lamotte (Aurum Aquaculture, 3-29-’94 AM Kit, code XX00153; Chestertown. Maryland, USA-21620), respectively.

It is evident from the present experiment that the captive breeding of the black carp can be operated successfully in commercial scale using Ovaprim in comparison to PG and LHRH.A2. The highest cost of Ovaprim over PG and LHRH.A2 may hinder its use by the commercial hatchery operators. However, higher cost could be offset by the performance of Ovaprim over PG or LHRH.A2 on viable oocyte development, fertilization rate, hatching rate and viable larvae production in black carp. On the other hand, LHRH.A2 should not be used at commercial scale considering operational time in relation to Ovaprim and cost in relation to PG. Considering the limitation of present study with a few brood (nine in total) and a single environmental condition, further extensive study would be needed to confirm the most suitable environmental condition and best dose of Ovaprim on the black carp breeding.
