Culturability Behaviour in Indica and Japonica Rice Varieties

Abstract/ Executive Summary:

An in vitro culturability (callus in duction and regeneration) study on indica and japonica rice (Oryza sativa L.) varieties were carried out using three explants types-mature seed, young panicle and immature embryo. Generally, compact, nodular and cream coloured calli showed better regeneration ability. On the other hand, loose, light yellowish and slightly watery calli were found less effective. The response in higher regeneration capability correlated to bigger size of compact calli. The regeneration ability of indica and japonica varieties interacted with explants type, variety and media combination. Immature embryo of BR5 showed better regeneration in both MS and K media compared to other indica and japonica varieties. Taipei309, used as experimental check. Higher number of regenerated green plants was obtained in MS medium form immature embryo of BR5 and BRRI dhan 29 and in K medium from BRRI dhan32. Media-explant interaction showed better regeneration in K medium from immature embryo. Similarly, explants-variety interaction also showed better regeneration from immature embryo of BR5. Therefore, BR5 could be used as a check variety instead in japonica variety, Taipei 309 for future study.

Objectives:

To observe the interaction of media to species in term of culturability and to develop the best regeneration system of test varieties, which could be used as check variety for indica rice

Methodology:

Different explants (eg mature seed, young panicle and immature embryo) of the five indica varieties-BR5, BRRI dhan28, BRRI dhan29, BRRI dhan 32, BRRI dhan40 and one japonica variety, Taipei309, were used as a control for comparative study on callus initiation and plant regeneration. Sterilized explant of different varieties were cultured on two different media namely MS with 2mg/l 2,4-D and K (modified MS medium). For regeneration MS basal salts having 1mg/l each of NAA and kinetin was used. The explants were cultured in callus induction media under dark incubation at 24±1^OC . After four weeks the calli developed from mature seeds were transferred to the regeneration medium. On the other hand, two weeks old calli derived from the young panicles and immature embryos were transferred to the regeneration medium. The cultures were incubated under continuous ligh for 4-5 weeks at 24±1^OC for shoot and root formation. The regenerated plants (green/albio) were transferred to hormone free MS medium for further growth and development. After acclimatized plants were then transferred to soil in pot and kept there until maturity. The experiments were set in completely randomized design (CRD) with 10-15 replications. Data on callusing and its size and structure were recorded before transferring to regeneration medium. Similarly, data on regeneration frequency and number of green and albino plant production were recorded after root shoot formation. Using the recorded data a descriptive analysis was done.
