Screening for Salinity Tolerance of Anther Derived Tissue Cultured and Breeding Lines at the Seedling Stage

Abstract/ Executive Summary:

The experiment was conducted to screen out salt tolerant anther cultured and breeding lines at seedling stage. Forty lines were screened at high saline condition using hydroponic system. Among them, only eight (8) lines namely BR7784-AC22-6-5-2-4, BR7784-AC28-6-2-2-3, BR7784-AC28-6-2-5-2, BR9782-AC5-1-3-1, BR7783-AC6-4-3-2-1-1, BR7784-AC22-6-1-3-3-2, BR7786-1B-2-1-3-3-3 and BR7786-1B-2-3-3-1-1 showed visual score 4 to 5 that is tolerant to moderately tolerant. However, other genotypes showed visual score 7 to 9 that is susceptible to highly susceptible at high saline condition.

Objectives:

To identify tolerant anther culture derived lines and breeding lines at seedling stage.

Methodology:

Thirty one Anther derived tissue cultured lines and nine breeding lines along with tolerant and susceptible check were considered for this experiment. Sprouted seeds were sown on Nylon net fitted with Styro-foam floating on Yoshida full strength culture solution according to Gregorio et al., 1997. Salinity stress @ 12 dS/m was applied 7 days after sowing. The culture solution was changed once a week throughout the experimental period. The pH and EC of the culture solution was checked daily and maintained at 5.0 and 12 dS/m respectively. In each tray FL478 and IR29 were used as a tolerant and susceptible check, respectively. When susceptible check IR29 died, visual score was made according to SES system and living plants were recorded. The experiment was conducted in RCB design with 2 replications.

Eight advanced lines were identified as tolerant to moderately tolerant to salt stress at the seedling stage. These genotypes could be used for future breeding program to develop new tolerant variety.
