Study on Biochemical Aspects of Submergence Tolerance

Abstract/ Executive Summary:

The experiment was conducted at Plant Physiology Division, Bangladesh Rice Research Institute to determine the change in dry weight, carbohydrate content, protein content and chlorophyll content during submergence period of tolerant and susceptible varieties. The dry weight of all varieties increased during the first nine days of submergence. After nine days all the varieties exhibited a decrease in dry weight. At ninth day after submergence dry matter of BR5, BR16 and FR13A were 107 gm/seedling. After 12 days after submergence dry matter of BR5, BR16 and FR13A were 34, 45 and 75.9 gm/seedling respectively. After 12 days of submergence carbohydrate content of FR13A had 1.98% followed by BR16 having 1.09%. BR5 had the lowest carbohydrate content of 0.30% after 12 days of submergence. In case of protein content, 12 days after submergence BR5, BR16 and FR13A had 12.25, 10.6 and 9.50 gm/seedling respectively. Chlorophyll content was higher at 3 days after submergence than 12 days after submergence. The highest chlorophyll was found in FR13A (1.13 mg/gm followed by BR16 (0.75 mg/gm) at twelve days after submergence.

Objectives:

To determine the change in dry weight, carbohydrate content, protein content and chlorophyll content during submergence period of tolerant and susceptible varieties.

Methodology:

BR5, BR16 and FR13A were tested in this experiment. Seedlings were grown in aluminum trays containing soil-sand culture mixed with nutrient solutions. Seedlings were grown for 10 days. The tray was allowed to complete submergence maintaining 70 cm water depth for 12 days. During submergence period, the water of the tank was made turbid twice daily and the light intensity in upper level (normal), mid-level (30 cm below the water surface) and lower level (75 cm below the water surface) of the tank were measured through light meter (LI-250). The water pH and temperature were also recorded. At 12 days after submergence, the water drained out from the submergence tank. Plants were collected from tray for taking necessary data.
