Disease Reactions and Variations in F3 Offspring of Eggplant for Resistant to Fruit Rot

Abstract/ Executive Summary:

The disease reactions and variations in yield contributing characters of eggplant were studied in F3 generation. F3 plants which derived from crossing with IPM-31(Katabegun WS) and commercial varieties (Dohazari G and Laffa S) were grown out of seeds collected from selfed F2s. The experimental populations were also evaluated for resistance and concerned yield characters. Inoculation of eggplant cultivars and F3 generation were done with Phomopsis vexans under confined field condition. Percent disease index (PDI) of leaf ranged from 0.00% to 15.18% and percent fruit infection from 0.00% to 49.25%. Among the three parents, two (Dohazari G and Laffa S) were found susceptible (S) and one parent IPM-31 showed resistant (R) reaction against P. vexans. All F3 plants showed resistant reaction.

Objectives:

1. The plants were evaluated for genotypic and phenotypic characters to see the segregating nature for resistance, to determine the performance of F3 generation of crosses.

Methodology:

During the period from September 2006 to March 2007, the field experiment was conducted in the plant pathology Field Laboratory, BAU, Mymensingh. The experiment was conducted during the winter season belonging to the Brahmaputra alluvial soil tract characterized by silty loam to sandy loam with a pH value of 6.2 (Ahmed et al., 1998). A Randomized Block Design with three replications was laid out for the experiment and the plots were fifteen (5x3). The plot size was 2.25 m x 3.75 m with 3 rows in each plot and 5 plants in each row. Both row to row and plant to pant distance were 75 cm. Seeds of the parental, F3 populations of the intervarietal crosses of eggplant, viz. (1) Dohazari G x IPM-31 (Katabegun WS), (2) Laffa S x IPM-31 were obtained from IPM Lab, Department of Plant Pathology, BAU, Mymensingh, Bangladesh. At the age of 25 days, seedlings were transplanted to the field from the net house. After transplanting, Irrigation and other Intercultural operations were done as and when necessary. In each plot of each cultivar/entry, inoculation of 5 plants was done at flowering stage and 5 plants at fruiting stage. Culturing P. vexans isolates and preparation of spore suspension were done. Seventy-mililitre spore suspension (5 x 106 spore/ml) was sprayed onto each plant Another 5 plants were kept uninoculated (control). The spraying was done at afternoon and inoculated plants were covered with moist transparent polythene sheet for 24 hours for ensuring better infection. The harvesting was continued from mid January, 2007 to 1st week of March, 2007. Two fruits per entry were allowed to ripe and seeds from them were collected for growing in the following year. The parameters (%Leaf infection, %Leaf Area Diseased, %flower infection, %fruit infection, % Fruit Area Diseased and lesion size) were used for observation and data collection after inoculation on the incidence/severity of fruit rot. Records on expression of symptom on leaf, flower and fruit were taken after inoculation at an interval of seven days up to 21 days. Infection was expressed in percentage. To calculate the percent leaf infection, total number of leaves inoculated and number of leaves infected was counted. By a measuring scale, lesion size and %LAD/FAD(PDI) were estimated and disease reaction (Resistant, Tolerant, Moderately Tolerant, Moderately Susceptible, Susceptible, Highly Susceptible ) was graded.

Achievement/Findings:

The F3 generations of eggplant which were derived from the crosses Laffa S x IPM-31 and Dohazari G x IPM-31 showed resistance against P. vexans might be due to the insertion of resistant gene from IPM-31 to offsprings. In F3 (Dohazari G x IPM-31), fruit shape segregated in 3:1(Round:Long) indicating the fruit shape (round), trait of IPM-31 is a dominant character.
