Characterization for Salinity Tolerance of BRRI Dhan28-saltol Introgression Lines at Seedling Stage

Abstract/ Executive Summary:

The experiment was conducted to know the tolerance gained by introgression of a major effect salinity tolerance QTL Saltol in to a Bangladeshi mega rice variety BRRI dhan28. Based on the measured traits, FL378 (donor parent) was ranked top for most of the traits except survivability and root weight. Among the tested introgression lines (NIL412, 434, 683, 618, 607, 448, 657 and 576) were shown better tolerance than the recipient parent BRRI dhan28, but this improvement was statistically insignificant. Despite the insignificant improvement, NIL412 showed 100% survivability for 2 weeks of salinity stress @12 dSm^-1. However, there are no such significant improvement was observed for the Na⁺ and K⁺ contents and their ratios in the shoots. Most of the lines showed increased Na⁺/K⁺ ratios compared to recipient parent. Some NILs (NIL204 to NIL17) perform poorer than sensitive check IR29, this is probably due to the small introgression at the target loci as well as poor vigor at seedling stage.

Objectives:

To find out the tolerance gained by the BRRI dhan28-Saltol introgression lines at seedling stage

Methodology:

Twenty four fixed BRRI dhan28-Saltol introgression lines having different sized introgressed segments at the target Saltol QTL region were tested along with donor parent (FL378), recipient parent (BRRI dhan28), original Saltol donor (Pokkali) and standard sensitive check IR29 in hydroponics culture at seedling stage with salinity @12 dSm^-1. The experiment was conducted in RCB design with 3 replications.The experiment was conducted in the Net house of Plant Physiology Division during the period December 2012 to January 2013. Sprouted seeds of all tested BR11-Saltol lines along with parents (BR11 and FL378) and standard tolerant & susceptible checks Pokkali and IR29 were sown in the Styrofoam sheets floating in plastic trays containing Yoshida’s culture solution (Yoshida et al. 1976). At 14 days of seedling age, NaCl was applied in the screening trays to attain the electrical conductivity (EC) at 6 dSm^-1 of the culture solution. Then the EC of the culture solution was increased 2 dS/ m in every 2^nd day and finally fixed @12 dSm^-1. The culture solution was changed once a week throughout the experimental period. The pH and EC of the culture solution was checked daily and maintained @ 5±0.5 and 12±0.5, respectively. The sensitivity of each seedling was scored when the sensitive check was almost died. The leaf damage scoring was done according to the scoring system described in IRRI’s Standard Evaluation System, 1996. Immediately after scoring, the data of seedling height and root lengths were recorded and then the seedlings were sampled for dry mass and mineral content (Na⁺ and K⁺ content of shoot and root).

Achievement/Findings:

Eight BRRI dhan28-Saltol NILs (NIL412, NIL434, NIL683, NIL618, NIL607, NIL448, NIL657 and NIL576) were found moderately tolerant in comparison with parent BRRI dhan28 as sensitive @ 12 dSm^-1 salinity stress at seedling stage.
