Abstract/ Executive Summary:

The viability of the stored germplasm was monitored from randomly selected germplasm in three seasons those were stored for three years and most of the tested germplasm possessed more than 90% germination . The germplasm accessions rejuvenated and stored during 2008-09 in short term were found to be more than 90% germination except seventy four (74) accession of which 49 in T Aman and 25 in Aus season. The 74 stored germplasm in short term which showed below 80% germination will be grown in the following season for regeneration.The germination percentages of the 5 test samples (testers) in the medium term and long term storage BRRI genebank. In 2005, as germination % were very low to medium, so testers were changed with new seed having 98 -99% germination. Among five testers, Hashikalmi, Nizersail, Patnai 23, Dharial which stored before eighty’s still possess good viability in the medium term conditions whereas Purbachi showed only 5% germination in mid term storage and 7% germination in long term genebank, respectively. It is also apparent from the table that the initial germination percent of newly restored test varieties were 98-99% when stored and in July 2009, percent of germination ranges from 99 to 98% except Purbachi (5% germination) in Mid term and 98 to 96% except Purbachi (7% germination) in long term during July 2009.

Objectives:

To check and monitor the germination (%) of the stored germplasm in the short, medium and long term storage.

Methodology:

One hundred and twenty seven accessions in Aus season, 200 accessions in T. Aman and 150 accessions in Boro season were checked randomly for viability test during 2008-09 in short term storage. Seed viability was monitored by testing germination percentage of the stored germplasm in short, medium and long term as well as testers in the medium and long term storage. Five tester varieties viz: Dharial, Hashikalmi, Nizersail, Patnai 23 and Purbachi possessing a seed viability range from very high to very low were used as testers in the medium and long term storage and viability was measured on every six month interval usually on July and December of every year for prediction of the viability of mid-term and long term storage germplasm. Silica gel was used to maintain the optimum moisture/relative humidity in the seed container. When the color of silica gel had been changed from blue to pinkish, then the silica gel were taken out and replaced by the fresh blue silica gel. The containers used for seed storage in medium and long term storage are hermetically sealed.
