Varietal Resistant in Rice Against Brown Planthopper, Nilaparvata Lugens (stal) in Bangladesh

Abstract/ Executive Summary:

A green house experiment was carried out to isolate resistant local and exotic rice varieties against the natural brown plant hopper (BPH) population, collected from four different locations of Bangladesh in presence of one resistant and one susceptible check. The screening test showed that Swarnalata and ARC 10550 were resistant like T27A but the other varieties were found susceptible. The susceptible varieties were always preferred by BPH nymphs. But a greater number of insects were always found to feed on Swarnalata than on any of the other resistant varieties.

Objectives:

The present experiment has been conducted to identify and develop the resistant rice varieties against BPH and to see whether biotypic variation exist in the natural BPH population in Bangladesh.

Methodology:

Gravid adult females of BPH were collected from infested rice fields at Cox's Bazar, Satkhira and Natore and were transported to the greenhouse of Bangladesh Rice Research Institute, Joydevpur for rearing. Another local population was also chosen from the BPH greenhouse to make four different populations in total for the present experiment. The collected insects were separately reared on 45-50 day old BR3 rice plants grown previously in earthen pots kept inside rearing cages (45×45×15 cm in size). The insects were allowed to lay eggs on the rice plants for 2 days. Then the plants were transferred in another cage to complete the hatching of eggs and development of new generations. In all cages new rice plants were supplied once every two days keeping the insects inside the cage for laying eggs on fresh plants. Only 2^nd/3^rd instars nymphs were used for the following tests. Screening of differential varieties: Eight rice varieties of which six differential (Mudgo ASD 7. Kuruhondarawala, Hotel Samba, ARC 10550 and Swarnalata) having different genes for resistance to different biotypes and two checks (TN1-susceptible and T27A-resistant to all the BPH biotypes) were used for the studies. Seeds were sown in soil contained a wooden flat (65 cm × 45 cm × 15 cm) in 10 cm ling lines and 5 cm apart. Each line represented a variety. The lines were completely randomized with 4 replications. Thus there were 32 lines in 4 rows in a flat. In total 4 flats with some varieties sown in the same design were used for 4 insect populations. The flats were sprinkled with water every day and they were transferred to galvanized iron trays (200 cm × 90 cm × 15 cm) after the seeds began to germinate. On 5^th days of germination seedlings were thinned leaving 25 plants per line. On 7^th day plants were infested with a large population of 2^nd/3^rd instars of BPH nymphs, so that each seedling had the opportunity of receiving approximately 5-7 nymphs in average. When around 90% of the seedlings of the susceptible check TN1 were found that after 5-7 days of infestation plant damage for each variety was recorded (based on visual damage) rating of 0-9 scale) according to the Standard Evaluation system (SEES) developed by IRRI (Heinrich et al. 1985). Preference of BPH for the differential varieties: Four flats with the same set of seeds. Seeded and infested in the same procedure as for the screening test, were prepared for the preference test. All the procedure for the preference test were the same as those for the screening test except than each flat was kept covered with a fine net. 100 cm × 70 cm × 65 cm in size after infestation and instead of taking plant damage scores, the number of the insects feeding on the plants were counted 6, 24 and 48 hours after infestation the insect counts were taken from the same to plants of each line every time.
