Activities of Some Enzymes-protecting Photo-oxidative Damage of Rice During Post Submergence Period

Abstract/ Executive Summary:

In low lying areas flash flood submergence is a chronic problem for T.Aman rice. The recovery ability of plants during post submergence period is an important character to compensate the loss due to flash flood submergence. Besides, there are some role of scavengers enzymes like SOD, Catalase, Ascorbate peroxidase (APX) in the submergence tolerance of rice. An experiment was conducted to determine the specific activities of SOD, Catalase and Ascorbate peroxidase (APX) during post submergence period in Plant Physiology Division, BRRI, Gazipur in 2000. SOD activity was the highest prior to the submergence irrespective of tolerant and non-tolerant varieties. SOD activity was increased remarkably after 21 hours after the submergence compared at 0 hour. Among the scavenger enzymes, SOD assay before submergence might be useful to identify submergence tolerant and non-tolerant varieties.

Objectives:

To determine the specific activities of SOD, Catalase and Ascorbate peroxidase (APX) during post submergence period

Methodology:

Four rice varieties namely FR13A, Hechua, BR5 and Chauolamagi were used. Sprouted seeds were sown in steel trays containing puddle soil fertilized with Urea, TSP, MP and Gypsum @ 3-2-2-2 g/tray. Each tray had 8 rows of seedlings with 2 rows of each variety. After 10 days, the seedlings were thinned to 20 seedlings per row. Seedlings were allowed to grow for 15 days. Fifteen-day old seedlings were submerged in cemented water tank. The depth of water was 80 cm above the soil level. Before submergence 2-3 rice plants were clipped off for enzymes assay at zero hour. After submergence for 7 days, the trays were transferred to the net house and samples were collected at 0 hour, 7 hour, 14 hour, 21 hour, and 28 hour exposure in the sunlight for enzyme assay.

Achievement/Findings:

In all the tested varieties the activity of SOD was the highest prior to the submergence. During post submergence period, the SOD activity of all the varieties was the lowest at 0 hour which was increased remarkably after 21 hours after the submergence. The enzymes like Catalase and APX exhibited the similar trend. It was remarkable that SOD, Catalase and APX difers in their activities before submergence among the varieties. The tolerant and non-tolerant varieties varied only SOD activity. This observation indicated that SOD was the only enzyme, which might relate with submergence tolerance of rice varieties. Submergence tolerant and non-tolerant varieties probably could be identified before submergence by SOD enzyme assay.
