Molecular Size of Osborne Rice Protein Fractions- 1994

Abstract/ Executive Summary:

Molecular sizes of different rice protein fractions were determined by SDS-Poly Acrylamide Gel Electrophoresis (SDS-PAGE). Glutelin, the major fraction of rice protein had at least 4 major molecular size proteins. There was variation in molecular size from variety to variety within a fraction. The average molecular size of the BR3 glutelin protein was bigger than that of glutelin protein of other varieties.

Objectives:

To isolate and fractionate the protein of the selected Bangladeshi rice varieties by polyacrylamide gel electrophoresis.

Methodology:

Four rice varieties BR3, Hbj IV, Rajashail and Binni were used for the study. Electrophoretic characterization of protein fraction had been pursued by SDS- polyacrylamide gel electrophoresis (SDS-PAGE).

Achievement/Findings:

SDS-page electrophoretic characterization of crude albumin extract showed one major band and 10 minor bands. Protein included in major band had molecular weight between 10,000 and 15,000 dalton only and constitutes more than 80% albumin of rajashail and Hbj tv and 34-50% albumin of BR3 and Binni. About 90% globulin of Rajashail had molecular weight between 15,000 and 25,000 dalton where as other varieties had only about 30% globulin within this molecular size. About 25% of globulin of BR3 and Binni rice had molecular weight between 40,000 and 50,000. About 50% of protein was included within this molecular size of all the varieties. Glutelin contained four major bands for all the varieties two had high MW and two had low MW.
