MS basal medium with 2mg/l2,4-D (MR medium)was found to be the best medium for callus induction. However, calli obtained from two different callus induction medium (MC and MR medium) were used for plant regeneration studies to examine the effect of callus induction medium on plant regeneration frequency.  Plant regeneration frequency was found to vary greatly depending on the callus induction medium, plant regeneration medium and the test genotypes. On an average, plant regeneration frequency was higher in RM2 and RM3 regeneration medium with calli obtained from MR medium (without casein hydrolysate) than those calli obtained from MC (with casein hydrolysate) induction medium. On the other hand, calli obtained from MC induction medium produced more green plant in RM1 and RM4 regeneration medium relative to MR induction medium. However, irrelevant of the effect of callus induction medium, percent pant regeneration was highest in RM1 regeneration medium followed by RM4, RM3 and RM2 medium respectively.

To achieve an efficient transgenic rice plant production, in vitro plant regeneration systems were evaluated using mature seed embryos as expants.

MS basal medium with 2mg/l2,4-D (MR medium)was found to be the best medium for callus induction. However, calli obtained from two different callus induction medium (MC and MR medium) were used for plant regeneration studies to examine the effect of callus induction medium on plant regeneration frequency. Moreover, an evaluation was undertaken to examine the performance of four different combinations of plant growth regulator (RM1, RM2, RM3 and RM4 medium) on green plant regeneration from mature embryo-derived calli of BRRI dhan29, BR11 and BR3.
